Mutations in this gene are associated with nonbullous congenital ichthyosiform erythroderma. [provided by RefSeq, Sep 2015]. * **UniProt Summary**: Non-heme iron-containing dioxygenase that catalyzes the stereo-specific peroxidation of free and esterified polyunsaturated fatty acids generating a spectrum of bioactive lipid mediators. Mainly converts arachidonic acid to (12R)- hydroperoxyeicosatetraenoic acid/(12R)-HPETE and minor stereoisomers. In the skin, acts upstream of ALOXE3 on the lineolate moiety of esterified omega-hydroxyacyl-sphingosine (EOS) ceramides to produce an epoxy-ketone derivative, a crucial step in the conjugation of omega-hydroxyceramide to membrane proteins. Therefore plays a crucial role in the synthesis of corneocytes lipid envelope and the establishment of the skin barrier to water loss. May also play a role in the regulation of the expression of airway mucins. {ECO:0000269|PubMed:21558561, ECO:0000269|PubMed:22441738, ECO:0000269|PubMed:9618483}. |PLAT| |Lipoxygenase| |linoleate 9S-lipoxygenase activity| |arachidonate 12-lipoxygenase activity| |hepoxilin metabolic process| |hepoxilin biosynthetic process| |positive regulation of mucus secretion| |regulation of mucus secretion| |oxidoreductase activity, acting on single donors with incorporation of molecular oxygen, incorporation of two atoms of oxygen| |lipoxygenase pathway| |linoleic acid metabolic process| |establishment of skin barrier| |regulation of water loss via skin| |long-chain fatty acid biosynthetic process| |unsaturated fatty acid biosynthetic process| |ceramide biosynthetic process| |arachidonic acid metabolic process| |multicellular organismal water homeostasis| |water homeostasis| |ceramide metabolic process| |protein lipidation| |sphingolipid biosynthetic process| |lipoprotein biosynthetic process| |unsaturated fatty acid metabolic process| |icosanoid metabolic process| |long-chain fatty acid metabolic process| |fatty acid biosynthetic process| |lipoprotein metabolic process| |membrane lipid biosynthetic process| |iron ion binding| |sphingolipid metabolic process| |fatty acid derivative metabolic process| |monocarboxylic acid biosynthetic process| |membrane lipid metabolic process| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |fatty acid metabolic process| |multicellular organismal homeostasis| |skin development| |positive regulation of secretion| |regulation of body fluid levels| |amide biosynthetic process| |monocarboxylic acid metabolic process| |positive regulation of MAPK cascade| |lipid biosynthetic process| |small molecule biosynthetic process| |regulation of MAPK cascade| |cellular amide metabolic process| |regulation of secretion| |carboxylic acid metabolic process| |cellular lipid metabolic process| |oxidation-reduction process| |positive regulation of transport| |oxoacid metabolic process| |positive regulation of protein phosphorylation| |positive regulation of intracellular signal transduction| |organic acid metabolic process| |positive regulation of phosphorylation| |chemical homeostasis| |positive regulation of phosphate metabolic process| |positive regulation of phosphorus metabolic process| |lipid metabolic process| |positive regulation of protein modification process| |organonitrogen compound biosynthetic process| |regulation of protein phosphorylation| |regulation of phosphorylation| |positive regulation of cellular protein metabolic process| |cellular nitrogen compound biosynthetic process| |homeostatic process| |positive regulation of signal transduction| |positive regulation of protein metabolic process| |cellular macromolecule biosynthetic process| |positive regulation of multicellular organismal process| |small molecule metabolic process| |macromolecule biosynthetic process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |regulation of protein modification process| |regulation of transport| |positive regulation of gene expression| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp310|2-Methoxyestradiol 0.55μM R07 exp310]]|1.74| |[[:results:exp210|LB-100 2μM R05 exp210]]|1.81| No correlation found to any other genes in chemogenomics.
