The protein encoded by this gene is involved in the activation of ARFs by accelerating replacement of bound GDP with GTP and is involved in Golgi transport. It contains a Sec7 domain, which may be responsible for its guanine-nucleotide exchange activity and also brefeldin A inhibition. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Promotes guanine-nucleotide exchange on ARF1 and ARF3 and to a lower extent on ARF5 and ARF6. Promotes the activation of ARF1/ARF5/ARF6 through replacement of GDP with GTP. Involved in the regulation of Golgi vesicular transport. Required for the integrity of the endosomal compartment. Involved in trafficking from the trans-Golgi network (TGN) to endosomes and is required for membrane association of the AP-1 complex and GGA1. Seems to be involved in recycling of the transferrin receptor from recycling endosomes to the plasma membrane. Probably is involved in the exit of GABA(A) receptors from the endoplasmic reticulum. Involved in constitutive release of tumor necrosis factor receptor 1 via exosome-like vesicles; the function seems to involve PKA and specifically PRKAR2B. Proposed to act as A kinase-anchoring protein (AKAP) and may mediate crosstalk between Arf and PKA pathways. {ECO:0000269|PubMed:12051703, ECO:0000269|PubMed:12571360, ECO:0000269|PubMed:15385626, ECO:0000269|PubMed:16477018, ECO:0000269|PubMed:17276987, ECO:0000269|PubMed:18625701, ECO:0000269|PubMed:20360857}. |Sec7 N| |Sec7| |DUF1981| |asymmetric synapse| |symmetric synapse| |axonemal microtubule| |receptor recycling| |GABA receptor binding| |regulation of ARF protein signal transduction| |ARF guanyl-nucleotide exchange factor activity| |protein kinase A regulatory subunit binding| |myosin binding| |Golgi to plasma membrane transport| |endosome organization| |positive regulation of tumor necrosis factor production| |positive regulation of tumor necrosis factor superfamily cytokine production| |vesicle-mediated transport to the plasma membrane| |post-Golgi vesicle-mediated transport| |guanyl-nucleotide exchange factor activity| |receptor metabolic process| |recycling endosome| |regulation of tumor necrosis factor production| |dendritic spine| |regulation of tumor necrosis factor superfamily cytokine production| |microtubule organizing center| |trans-Golgi network| |regulation of Ras protein signal transduction| |cytoplasmic vesicle| |vesicle organization| |regulation of small GTPase mediated signal transduction| |Golgi vesicle transport| |endomembrane system organization| |positive regulation of cytokine production| |cell junction| |Golgi membrane| |regulation of cytokine production| |perinuclear region of cytoplasm| |exocytosis| |secretion by cell| |export from cell| |secretion| |protein transport| |peptide transport| |amide transport| |establishment of protein localization| |intracellular signal transduction| |positive regulation of multicellular organismal process| |regulation of intracellular signal transduction| |nitrogen compound transport| |vesicle-mediated transport| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp434|Vemurafenib 6.6μM R08 exp434]]|-2.57| |[[:results:exp68|Clomiphene 4.4μM R02 exp68]]|-2.24| |[[:results:exp89|Vemurafenib 6.6μM R02 exp89]]|-2.14| |[[:results:exp287|HMS-I2 5μM R06 exp287]]|-1.92| |[[:results:exp53|Suberoylanilide-Hydroxamic-Acid 0.02μM R01 exp53]]|-1.7| |[[:results:exp346|CoCl2 18μM R07 exp346]]|1.75| |[[:results:exp320|ABT-702 5μM plus CoCl2 18μM R07 exp320]]|1.96| |[[:results:exp36|TRAIL 50ng/ml R00 exp36]]|2.13| No correlation found to any other genes in chemogenomics.
