This gene product catalyzes the formation of the glycosaminoglycan-protein linkage by way of a glucuronyl transfer reaction in the final step of the biosynthesis of the linkage region of proteoglycans. A pseudogene of this gene has been identified on chromosome 3. [provided by RefSeq, Dec 2013]. * **UniProt Summary**: Glycosaminoglycans biosynthesis (PubMed:25893793). Involved in forming the linkage tetrasaccharide present in heparan sulfate and chondroitin sulfate. Transfers a glucuronic acid moiety from the uridine diphosphate-glucuronic acid (UDP-GlcUA) to the common linkage region trisaccharide Gal-beta-1,3-Gal-beta-1,4- Xyl covalently bound to a Ser residue at the glycosaminylglycan attachment site of proteoglycans. Can also play a role in the biosynthesis of l2/HNK-1 carbohydrate epitope on glycoproteins. Shows strict specificity for Gal-beta-1,3-Gal-beta-1,4-Xyl, exhibiting negligible incorporation into other galactoside substrates including Galbeta1-3Gal beta1-O-benzyl, Galbeta1- 4GlcNAc and Galbeta1-4Glc. Stimulates 2-phosphoxylose phosphatase activity of PXYLP1 in presence of uridine diphosphate-glucuronic acid (UDP-GlcUA) during completion of linkage region formation (PubMed:24425863). {ECO:0000269|PubMed:24425863, ECO:0000269|PubMed:25893793}. |Glyco transf 43| |galactosylgalactosylxylosylprotein 3-beta-glucuronosyltransferase activity| |protein phosphatase activator activity| |dermatan sulfate proteoglycan biosynthetic process| |dermatan sulfate proteoglycan metabolic process| |glucuronosyltransferase activity| |heparan sulfate proteoglycan biosynthetic process| |chondroitin sulfate proteoglycan biosynthetic process| |heparan sulfate proteoglycan metabolic process| |chondroitin sulfate proteoglycan metabolic process| |cis-Golgi network| |proteoglycan biosynthetic process| |proteoglycan metabolic process| |glycosaminoglycan biosynthetic process| |aminoglycan biosynthetic process| |regulation of phosphoprotein phosphatase activity| |regulation of protein dephosphorylation| |positive regulation of intracellular protein transport| |glycosaminoglycan metabolic process| |aminoglycan metabolic process| |regulation of phosphatase activity| |positive regulation of intracellular transport| |regulation of dephosphorylation| |regulation of intracellular protein transport| |protein glycosylation| |macromolecule glycosylation| |glycosylation| |glycoprotein biosynthetic process| |positive regulation of cellular protein localization| |regulation of intracellular transport| |glycoprotein metabolic process| |positive regulation of protein transport| |positive regulation of establishment of protein localization| |regulation of cellular protein localization| |Golgi membrane| |carbohydrate derivative biosynthetic process| |regulation of protein transport| |regulation of peptide transport| |regulation of establishment of protein localization| |regulation of cellular localization| |Golgi apparatus| |positive regulation of transport| |regulation of protein localization| |carbohydrate derivative metabolic process| |regulation of hydrolase activity| |organonitrogen compound biosynthetic process| |positive regulation of catalytic activity| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |positive regulation of molecular function| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| |regulation of transport| |membrane| \\ === CRISPR Data === ^Screen^Score^ |Hinokiflavone 12μM R08 exp487|-1.78| |CCT251545 20μM R07 exp326|-1.73| |Pyridostatin 4μM R05 exp240|2.75| ^Gene^Correlation^ |PAPSS1|0.426| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 6607 * **Expression level (log2 read counts)**: 5.11
