The protein forms a complex with p53 and negatively regulates p53 transcription, and functions as a tumor suppressor and cell cycle regulator. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Mar 2010]. * **UniProt Summary**: Controls V(D)J recombination during T-cell development by repressing T-cell receptor (TCR) beta enhancer function. Binds to scaffold/matrix attachment region beta (S/MARbeta), an ATC-rich DNA sequence located upstream of the TCR beta enhancer. Represses cyclin D1 transcription by recruiting HDAC1 to its promoter, thereby diminishing H3K9ac, H3S10ph and H4K8ac levels. Promotes TP53 'Ser-15' phosphorylation and nuclear accumulation, which causes cell cycle arrest (By similarity). {ECO:0000250, ECO:0000269|PubMed:16166625}. |BEN| |p53 binding| |negative regulation of protein catabolic process| |protein localization to nucleus| |regulation of signal transduction by p53 class mediator| |nuclear body| |negative regulation of catabolic process| |regulation of protein catabolic process| |chromatin organization| |protein localization to organelle| |regulation of catabolic process| |chromosome organization| |negative regulation of protein metabolic process| |cell cycle| |DNA binding| |positive regulation of transcription, DNA-templated| |cellular protein localization| |cellular macromolecule localization| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |positive regulation of RNA metabolic process| |regulation of intracellular signal transduction| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Ethanol 0.01 R05 exp234|1.83| |Tunicamycin 0.04μM R07 exp418|1.86| |Thalidomide 20μM R07 exp406|1.94| ^Gene^Correlation^ |RRM1|0.428| Global Fraction of Cell Lines Where Essential: 215/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|0/1| |bile duct|7/28| |blood|9/28| |bone|14/25| |breast|13/33| |central nervous system|19/56| |cervix|1/4| |colorectal|2/17| |esophagus|5/13| |fibroblast|1/1| |gastric|2/15| |kidney|4/21| |liver|10/20| |lung|26/75| |lymphocyte|2/14| |ovary|9/26| |pancreas|7/24| |peripheral nervous system|8/16| |plasma cell|5/15| |prostate|1/1| |skin|3/24| |soft tissue|1/7| |thyroid|1/2| |upper aerodigestive|2/22| |urinary tract|7/29| |uterus|1/5| == Essentiality in NALM6 == * **Essentiality Rank**: 2526 * **Expression level (log2 read counts)**: 3.7
