The encoded protein is an actin-binding protein that is activated by the serine/threonine kinase Akt and plays a role in cytoskeleton remodeling and cell migration. The encoded protein also enhances Akt signaling by mediating phosphoinositide 3-kinase (PI3K)-dependent activation of Akt by growth factor receptor tyrosine kinases and G protein-coupled receptors. Increased expression of this gene and phosphorylation of the encoded protein may play a role in cancer metastasis. Alternatively spliced transcript variants encoding multiple isoforms have been observed for this gene. [provided by RefSeq, Dec 2011]. * **UniProt Summary**: Plays a role as a key modulator of the AKT-mTOR signaling pathway controlling the tempo of the process of newborn neurons integration during adult neurogenesis, including correct neuron positioning, dendritic development and synapse formation (By similarity). Enhances phosphoinositide 3-kinase (PI3K)- dependent phosphorylation and kinase activity of AKT1/PKB, but does not possess kinase activity itself (By similarity). Phosphorylation of AKT1/PKB thereby induces the phosphorylation of downstream effectors GSK3 and FOXO1/FKHR, and regulates DNA replication and cell proliferation (By similarity). Essential for the integrity of the actin cytoskeleton and for cell migration (PubMed:16139227). Required for formation of actin stress fibers and lamellipodia (PubMed:15882442). May be involved in membrane sorting in the early endosome (PubMed:15882442). Plays a role in ciliogenesis and cilium morphology and positioning and this may partly be through regulation of the localization of scaffolding protein CROCC/Rootletin (PubMed:27623382). {ECO:0000250|UniProtKB:Q5SNZ0, ECO:0000269|PubMed:15882442, ECO:0000269|PubMed:16139227, ECO:0000269|PubMed:27623382}. |HOOK| |COPI-coated Golgi to ER transport vesicle| |G-protein gamma-subunit binding| |positive regulation of protein localization to cilium| |regulation of protein localization to cilium| |protein kinase B binding| |positive regulation of cilium assembly| |TOR signaling| |dynein light intermediate chain binding| |activation of protein kinase B activity| |lamellipodium assembly| |lamellipodium organization| |cytoplasmic microtubule organization| |positive regulation of organelle assembly| |regulation of cilium assembly| |phosphatidylinositol binding| |positive regulation of plasma membrane bounded cell projection assembly| |regulation of DNA replication| |ciliary basal body| |centriole| |cytoskeleton-dependent intracellular transport| |lamellipodium| |regulation of plasma membrane bounded cell projection assembly| |regulation of cell projection assembly| |DNA replication| |regulation of organelle assembly| |microtubule binding| |cytoplasmic vesicle| |actin binding| |positive regulation of cellular protein localization| |activation of protein kinase activity| |regulation of actin cytoskeleton organization| |positive regulation of cell projection organization| |regulation of actin filament-based process| |plasma membrane bounded cell projection assembly| |supramolecular fiber organization| |cell projection assembly| |microtubule cytoskeleton organization| |centrosome| |regulation of neuron projection development| |positive regulation of cellular component biogenesis| |positive regulation of protein kinase activity| |regulation of cellular protein localization| |regulation of cytoskeleton organization| |positive regulation of kinase activity| |positive regulation of organelle organization| |positive regulation of transferase activity| |regulation of neuron differentiation| |microtubule-based process| |regulation of plasma membrane bounded cell projection organization| |regulation of cell projection organization| |regulation of protein kinase activity| |regulation of neurogenesis| |membrane organization| |regulation of kinase activity| |protein homodimerization activity| |regulation of cellular localization| |regulation of nervous system development| |regulation of cell development| |regulation of cellular component biogenesis| |cell migration| |regulation of transferase activity| |Golgi apparatus| |endoplasmic reticulum| |positive regulation of protein phosphorylation| |regulation of protein localization| |positive regulation of phosphorylation| |localization of cell| |cell motility| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |positive regulation of phosphate metabolic process| |positive regulation of phosphorus metabolic process| |cell projection organization| |positive regulation of cellular component organization| |positive regulation of protein modification process| |regulation of organelle organization| |locomotion| |positive regulation of catalytic activity| |regulation of protein phosphorylation| |intracellular transport| |generation of neurons| |movement of cell or subcellular component| |regulation of phosphorylation| |positive regulation of cellular protein metabolic process| |regulation of cell population proliferation| |neurogenesis| |intracellular signal transduction| |positive regulation of protein metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |positive regulation of molecular function| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of cell differentiation| |establishment of localization in cell| |regulation of protein modification process| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp306|Rapamycin 2μM R07 exp306]]|-1.78| |[[:results:exp208|Vinblastine 0.015μM R05 exp208]]|1.81| No correlation found to any other genes in chemogenomics.
