This family regulates CREB-dependent gene transcription in a phosphorylation-independent manner and may be selective for cAMP-responsive genes. The protein encoded by this gene may induce mitochondrial biogenesis and attenuate catecholamine signaling in adipose tissue. A translocation event between this gene and Notch coactivator mastermind-like gene 2, which results in a fusion protein, has been reported in mucoepidermoid carcinomas. Alternative splicing results in multiple transcript variants that encode different protein isoforms. [provided by RefSeq, Jul 2012]. * **UniProt Summary**: Transcriptional coactivator for CREB1 which activates transcription through both consensus and variant cAMP response element (CRE) sites. Acts as a coactivator, in the SIK/TORC signaling pathway, being active when dephosphorylated and acts independently of CREB1 'Ser-133' phosphorylation. Enhances the interaction of CREB1 with TAF4. Regulates the expression of specific CREB-activated genes such as the steroidogenic gene, StAR. Potent coactivator of PPARGC1A and inducer of mitochondrial biogenesis in muscle cells. Also coactivator for TAX activation of the human T-cell leukemia virus type 1 (HTLV-1) long terminal repeats (LTR). {ECO:0000269|PubMed:14506290, ECO:0000269|PubMed:15454081, ECO:0000269|PubMed:15466468, ECO:0000269|PubMed:16817901, ECO:0000269|PubMed:16980408, ECO:0000269|PubMed:17210223, ECO:0000269|PubMed:17644518}. |TORC N| |TORC M| |TORC C| |cAMP response element binding protein binding| |positive regulation of CREB transcription factor activity| |negative regulation of cAMP-mediated signaling| |negative regulation of lipid catabolic process| |energy homeostasis| |regulation of cAMP-mediated signaling| |macrophage activation| |regulation of lipid catabolic process| |negative regulation of lipid metabolic process| |protein homotetramerization| |protein tetramerization| |positive regulation of DNA-binding transcription factor activity| |negative regulation of catabolic process| |multicellular organismal homeostasis| |protein homooligomerization| |regulation of lipid metabolic process| |regulation of DNA-binding transcription factor activity| |negative regulation of intracellular signal transduction| |protein complex oligomerization| |myeloid leukocyte activation| |viral process| |symbiotic process| |interspecies interaction between organisms| |leukocyte activation| |regulation of catabolic process| |cell activation| |positive regulation of transcription by RNA polymerase II| |negative regulation of signal transduction| |negative regulation of cell communication| |negative regulation of signaling| |positive regulation of transcription, DNA-templated| |protein-containing complex assembly| |negative regulation of response to stimulus| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |homeostatic process| |positive regulation of RNA metabolic process| |positive regulation of molecular function| |regulation of intracellular signal transduction| |protein-containing complex subunit organization| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Geldanamycin 0.015 to 0.05μM on day4 R07 exp317|-2.27| |IU1-47 25μM R04 exp182|-1.97| |SGC2043 10μM R03 exp152|-1.78| |Ibrutinib 10μM R08 exp442|-1.77| |ETC-159 50μM R08 exp480|-1.7| ^Gene^Correlation^ |RRM1|0.445| Global Fraction of Cell Lines Where Essential: 3/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|1/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 7082 * **Expression level (log2 read counts)**: 4.37
