The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF- 2:GTP:methionyl-tRNAi and eIF-5 to form the 43S pre-initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation. The eIF-3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed:25849773). {ECO:0000269|PubMed:25849773, ECO:0000269|PubMed:27462815}. |eIF3 subunit| |eukaryotic 48S preinitiation complex| |formation of cytoplasmic translation initiation complex| |eukaryotic translation initiation factor 3 complex| |eukaryotic 43S preinitiation complex| |cytoplasmic translational initiation| |translation initiation factor activity| |cytoplasmic translation| |translational initiation| |ribonucleoprotein complex assembly| |ribonucleoprotein complex subunit organization| |translation| |peptide biosynthetic process| |ribonucleoprotein complex biogenesis| |amide biosynthetic process| |peptide metabolic process| |cellular amide metabolic process| |cellular protein-containing complex assembly| |organonitrogen compound biosynthetic process| |protein-containing complex assembly| |cellular nitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |protein-containing complex subunit organization| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Rapamycin 1μM R00 exp29|-1.82| |UM0129480 7μM R02 exp85|-1.77| |NMS-873 0.07μM R07 exp380|1.81| |Cytochalasin-B 5μM R07 exp349|1.88| |Nicotinate 1000μM R03 exp140|1.92| |UNC1999 2μM R03 exp155|2| |IU1-C 25μM R04 exp183|2.06| |MKC9989 10μM R03 exp133|2.11| |GSK-LSD1 10μM R03 exp123|2.14| |BI-6727 0.0125μM R03 exp97|2.17| |CR131-b 0.005μM R08 exp474|2.3| |BI-2536 0.0042μM R03 exp95|2.92| ^Gene^Correlation^ |HGC6.3|0.498| Global Fraction of Cell Lines Where Essential: 3/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|1/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1403 * **Expression level (log2 read counts)**: 7.04
