The vitamin K-dependent protein substrates have a propeptide that binds the enzyme, with carbon dioxide, dioxide, and reduced vitamin K acting as co-substrates. Vitamin K-dependent proteins affect a number of physiologic processes including blood coagulation, prevention of vascular calcification, and inflammation. Allelic variants of this gene have been associated with pseudoxanthoma elasticum-like disorder with associated multiple coagulation factor deficiency. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Aug 2015]. * **UniProt Summary**: Mediates the vitamin K-dependent carboxylation of glutamate residues to calcium-binding gamma-carboxyglutamate (Gla) residues with the concomitant conversion of the reduced hydroquinone form of vitamin K to vitamin K epoxide. {ECO:0000269|PubMed:17073445}. |VKG Carbox| |gamma-glutamyl carboxylase activity| |peptidyl-glutamic acid carboxylation| |protein carboxylation| |peptidyl-glutamic acid modification| |blood coagulation| |coagulation| |hemostasis| |wound healing| |regulation of body fluid levels| |response to wounding| |peptidyl-amino acid modification| |endoplasmic reticulum membrane| |membrane| \\ === CRISPR Data === ^Screen^Score^ |Suberoylanilide-Hydroxamic-Acid 0.02μM R01 exp53|-2.22| |BML-284 0.09μM R08 exp460|-1.92| |Amiloride 100μM R08 exp447|1.71| |PU-H71 1μM R08 exp515|1.77| |Nocodazole 0.1μM R03 exp94|1.82| |Cannabidiol 20μM R08 exp466|2.13| ^Gene^Correlation^ |RRM1|0.497| Global Fraction of Cell Lines Where Essential: 1/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 16216 * **Expression level (log2 read counts)**: 6.64
