This gene encodes a growth factor receptor-binding protein that interacts with insulin receptors and insulin-like growth-factor receptors. This protein likely has an inhibitory effect on receptor tyrosine kinase signaling and, in particular, on insulin receptor signaling. This gene may play a role in signaling pathways that regulate growth and metabolism. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2014]. * **UniProt Summary**: Adapter protein which modulates coupling of cell surface receptor kinases with specific signaling pathways. Binds to, and suppresses signals from, the activated insulin receptor (INSR). Potent inhibitor of insulin-stimulated MAPK3 phosphorylation. Plays a critical role regulating PDPK1 membrane translocation in response to insulin stimulation and serves as an adapter protein to recruit PDPK1 to activated insulin receptor, thus promoting PKB/AKT1 phosphorylation and transduction of the insulin signal. {ECO:0000269|PubMed:15210700, ECO:0000269|PubMed:19648926}. |PH| |RA| |SH2| |BPS| |negative regulation of insulin receptor signaling pathway| |negative regulation of cellular response to insulin stimulus| |SH3/SH2 adaptor activity| |receptor tyrosine kinase binding| |regulation of insulin receptor signaling pathway| |regulation of cellular response to insulin stimulus| |insulin receptor signaling pathway| |cellular response to insulin stimulus| |endosome membrane| |response to insulin| |cellular response to peptide hormone stimulus| |cellular response to peptide| |leukocyte migration| |response to peptide hormone| |response to peptide| |transmembrane receptor protein tyrosine kinase signaling pathway| |cellular response to organonitrogen compound| |cellular response to hormone stimulus| |cellular response to nitrogen compound| |intracellular membrane-bounded organelle| |enzyme linked receptor protein signaling pathway| |protein homodimerization activity| |response to hormone| |cell migration| |response to organonitrogen compound| |cellular response to oxygen-containing compound| |identical protein binding| |cell motility| |localization of cell| |response to nitrogen compound| |cellular response to endogenous stimulus| |negative regulation of signal transduction| |locomotion| |negative regulation of cell communication| |negative regulation of signaling| |response to endogenous stimulus| |movement of cell or subcellular component| |response to oxygen-containing compound| |negative regulation of response to stimulus| |positive regulation of signal transduction| |positive regulation of cell communication| |positive regulation of signaling| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp475|CyclicAMP 200μM R08 exp475]]|-2| |[[:results:exp525|Sulforaphane 9μM R08 exp525]]|-1.79| |[[:results:exp456|Benzoate 20000μM R08 exp456]]|1.71| |[[:results:exp151|SGC0946 7μM R03 exp151]]|1.97| No correlation found to any other genes in chemogenomics.
