The encoded protein hydrolyzes inosine triphosphate and deoxyinosine triphosphate to the monophosphate nucleotide and diphosphate. This protein, which is a member of the HAM1 NTPase protein family, is found in the cytoplasm and acts as a homodimer. Defects in the encoded protein can result in inosine triphosphate pyrophosphorylase deficiency which causes an accumulation of ITP in red blood cells. Alternate splicing results in multiple transcript variants. [provided by RefSeq, Jun 2012]. * **UniProt Summary**: Pyrophosphatase that hydrolyzes the non-canonical purine nucleotides inosine triphosphate (ITP), deoxyinosine triphosphate (dITP) as well as 2'-deoxy-N-6-hydroxylaminopurine triposphate (dHAPTP) and xanthosine 5'-triphosphate (XTP) to their respective monophosphate derivatives. The enzyme does not distinguish between the deoxy- and ribose forms. Probably excludes non-canonical purines from RNA and DNA precursor pools, thus preventing their incorporation into RNA and DNA and avoiding chromosomal lesions. {ECO:0000255|HAMAP-Rule:MF_03148, ECO:0000269|PubMed:17090528}. |Ham1p like| |ITP catabolic process| |purine ribonucleoside triphosphate catabolic process| |ribonucleoside triphosphate catabolic process| |dITP diphosphatase activity| |ITP metabolic process| |purine nucleoside triphosphate catabolic process| |nucleoside-triphosphate diphosphatase activity| |NADH pyrophosphatase activity| |deoxyribonucleoside triphosphate catabolic process| |nucleoside triphosphate catabolic process| |deoxyribonucleoside triphosphate metabolic process| |purine ribonucleotide catabolic process| |ribonucleotide catabolic process| |purine nucleotide catabolic process| |purine-containing compound catabolic process| |purine ribonucleoside triphosphate metabolic process| |nucleotide binding| |nucleotide catabolic process| |ribonucleoside triphosphate metabolic process| |purine nucleoside triphosphate metabolic process| |nucleoside phosphate catabolic process| |nucleoside triphosphate metabolic process| |nucleobase-containing small molecule biosynthetic process| |organophosphate catabolic process| |carbohydrate derivative catabolic process| |purine ribonucleotide metabolic process| |ribonucleotide metabolic process| |purine nucleotide metabolic process| |ribose phosphate metabolic process| |purine-containing compound metabolic process| |nucleobase-containing compound catabolic process| |heterocycle catabolic process| |cellular nitrogen compound catabolic process| |aromatic compound catabolic process| |nucleotide metabolic process| |nucleoside phosphate metabolic process| |organic cyclic compound catabolic process| |nucleobase-containing small molecule metabolic process| |small molecule biosynthetic process| |organophosphate metabolic process| |carbohydrate derivative metabolic process| |organonitrogen compound catabolic process| |chromosome organization| |identical protein binding| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |organic cyclic compound biosynthetic process| |cellular nitrogen compound biosynthetic process| |small molecule metabolic process| |organic substance catabolic process| |cellular catabolic process| \\ === CRISPR Data === ^Screen^Score^ |Erlotinib 10μM R05 exp216|-3.04| |Rucaparib 6.5μM R08 exp520|-2.62| |Olaparib 4μM R08 exp512|-2.54| |Hypoxia 5%O2 R04 exp191|-2.01| |Calcipotriol 5μM R08 exp464|-1.99| |Talazoparib 0.006μM R06 exp299|-1.96| |5-Fluorouracil 2μM R00 exp1|-1.76| |LB-100 2μM R05 exp210|-1.73| |ABT-702 5μM R07 exp318|-1.7| |Sulforaphane 9μM R08 exp526 no dilution day6|2.02| |MLN-4924 2μM R00 exp22|2.19| ^Gene^Correlation^ |RNASEH2B|0.484| |ELP2|0.443| |EIF1AX|0.412| |URM1|0.4| Global Fraction of Cell Lines Where Essential: 3/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|1/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|1/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 10676 * **Expression level (log2 read counts)**: 5.12
