The encoded preproprotein present in plasma as a non-covalent complex with high molecular weight kininogen undergoes proteolytic processing mediated by activated coagulation factor XII to generate a disulfide-linked, heterodimeric serine protease comprised of heavy and light chains. Certain mutations in this gene cause prekallikrein deficiency. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Jan 2016]. * **UniProt Summary**: The enzyme cleaves Lys-Arg and Arg-Ser bonds. It activates, in a reciprocal reaction, factor XII after its binding to a negatively charged surface. It also releases bradykinin from HMW kininogen and may also play a role in the renin-angiotensin system by converting prorenin into renin. |PAN 1| |Trypsin| |Factor XII activation| |kinin cascade| |plasma kallikrein-kinin cascade| |positive regulation of fibrinolysis| |plasminogen activation| |regulation of fibrinolysis| |blood coagulation, intrinsic pathway| |fibrinolysis| |positive regulation of protein processing| |positive regulation of protein maturation| |protein activation cascade| |blood coagulation, fibrin clot formation| |positive regulation of acute inflammatory response| |zymogen activation| |regulation of acute inflammatory response| |negative regulation of blood coagulation| |negative regulation of hemostasis| |negative regulation of coagulation| |extracellular matrix disassembly| |regulation of protein processing| |regulation of protein maturation| |acute inflammatory response| |negative regulation of wound healing| |regulation of blood coagulation| |regulation of hemostasis| |regulation of coagulation| |negative regulation of response to wounding| |regulation of wound healing| |positive regulation of inflammatory response| |protein processing| |serine-type endopeptidase activity| |regulation of response to wounding| |protein maturation| |blood coagulation| |coagulation| |hemostasis| |regulation of inflammatory response| |extracellular matrix organization| |positive regulation of proteolysis| |negative regulation of response to external stimulus| |extracellular structure organization| |cellular component disassembly| |positive regulation of defense response| |wound healing| |regulation of body fluid levels| |inflammatory response| |response to wounding| |positive regulation of response to external stimulus| |regulation of proteolysis| |regulation of defense response| |regulation of response to external stimulus| |negative regulation of multicellular organismal process| |proteolysis| |defense response| |regulation of response to stress| |extracellular space| |positive regulation of cellular protein metabolic process| |negative regulation of response to stimulus| |positive regulation of protein metabolic process| |extracellular region| |positive regulation of gene expression| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp5|Alpha-Amanitin 0.5μM R00 exp5]]|-2.08| No correlation found to any other genes in chemogenomics.
