The encoded protein functions in DNA replication, recombination, and the base excision repair process. Mutations in this gene that lead to DNA ligase I deficiency result in immunodeficiency and increased sensitivity to DNA-damaging agents. Disruption of this gene may also be associated with a variety of cancers. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Jan 2014]. * **UniProt Summary**: DNA ligase that seals nicks in double-stranded DNA during DNA replication, DNA recombination and DNA repair. |DNA ligase A M| |DNA ligase A N| |DNA ligase A C| |Okazaki fragment processing involved in mitotic DNA replication| |DNA replication, Okazaki fragment processing| |DNA ligase activity| |DNA ligase (ATP) activity| |lagging strand elongation| |DNA ligation involved in DNA repair| |mitotic DNA replication| |DNA ligation| |V(D)J recombination| |DNA strand elongation involved in DNA replication| |DNA strand elongation| |nucleotide-excision repair, DNA gap filling| |mismatch repair| |somatic diversification of immune receptors via germline recombination within a single locus| |somatic cell DNA recombination| |base-excision repair| |nuclear DNA replication| |cell cycle DNA replication| |somatic diversification of immune receptors| |transcription-coupled nucleotide-excision repair| |DNA biosynthetic process| |nucleotide-excision repair| |DNA-dependent DNA replication| |DNA replication| |DNA recombination| |cell division| |DNA repair| |mitotic cell cycle process| |immune system development| |mitotic cell cycle| |intracellular membrane-bounded organelle| |DNA metabolic process| |cellular response to DNA damage stimulus| |cell cycle process| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |mitochondrion| |organic cyclic compound biosynthetic process| |cell cycle| |DNA binding| |ATP binding| |cellular nitrogen compound biosynthetic process| |cellular response to stress| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Nocodazole 0.2μM R02 exp64|-2.28| |Latrunculin-B 10μM R07 exp374|-1.84| |Vinblastine 0.015μM R05 exp208|-1.83| |A-769662 20μM R07 exp331|-1.76| |HMS-I1 10μM R06 exp256|-1.73| |Phenformin 20μM R05 exp212|1.7| |Epothilone-B 0.0015μM R05 exp231|1.74| |LJH685 5μM R02 exp73|1.74| |Dorsomorphin 5μM R07 exp357|1.75| |Etoposide 0.1μM R05 exp198|1.75| |Glyphosate 1000μM R06 exp283|1.77| |Tubastatin-A 2.5μM R07 exp417|1.84| |WYE-354 6μM R02 exp90|1.93| |2-Methoxyestradiol 0.55μM R07 exp310|2.02| |39°C R06 exp303|2.04| |Adefovir 20μM R07 exp332|2.08| |Deguelin 0.15μM R05 exp209|2.32| |Geldanamycin 0.01μM R05 exp235|2.41| |Zebularine 20μM R07 exp423|2.53| |LB-100 4.1μM R02 exp72|2.67| |BN82002 4μM R07 exp340|2.8| |FK-506 30μM R07 exp359|2.86| |Demecolcine 0.03μM R05 exp228|2.9| |Clomiphene 4.4μM R02 exp68|2.98| ^Gene^Correlation^ |EME1|0.435| |PIWIL1|0.416| |BRD8|0.414| Global Fraction of Cell Lines Where Essential: 4/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|2/26| |breast|1/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|1/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 381 * **Expression level (log2 read counts)**: 6.95
