Mutations at this locus have been associated with autosomal dominant thrombocytopenia, also known as thrombocytopenia-2. Alternatively spliced transcript variants have been described for this locus. [provided by RefSeq, Feb 2010]. * **UniProt Summary**: Serine/threonine kinase that plays a key role in M phase by acting as a regulator of mitosis entry and maintenance. Acts by promoting the inactivation of protein phosphatase 2A (PP2A) during M phase: does not directly inhibit PP2A but acts by mediating phosphorylation and subsequent activation of ARPP19 and ENSA at 'Ser-62' and 'Ser-67', respectively. ARPP19 and ENSA are phosphatase inhibitors that specifically inhibit the PPP2R2D (PR55-delta) subunit of PP2A. Inactivation of PP2A during M phase is essential to keep cyclin-B1-CDK1 activity high. Following DNA damage, it is also involved in checkpoint recovery by being inhibited. Phosphorylates histone protein in vitro; however such activity is unsure in vivo. May be involved in megakaryocyte differentiation. {ECO:0000269|PubMed:12890928, ECO:0000269|PubMed:19680222, ECO:0000269|PubMed:19793917, ECO:0000269|PubMed:20538976, ECO:0000269|PubMed:20818157}. |Pkinase| |Pkinase Tyr| |female meiosis II| |meiosis II cell cycle process| |meiosis II| |positive regulation of ubiquitin protein ligase activity| |regulation of ubiquitin protein ligase activity| |female meiotic nuclear division| |positive regulation of ubiquitin-protein transferase activity| |protein phosphatase 2A binding| |regulation of ubiquitin-protein transferase activity| |cleavage furrow| |negative regulation of phosphoprotein phosphatase activity| |negative regulation of protein dephosphorylation| |kinase activity| |negative regulation of phosphatase activity| |negative regulation of dephosphorylation| |positive regulation of protein ubiquitination| |regulation of phosphoprotein phosphatase activity| |female gamete generation| |G2/M transition of mitotic cell cycle| |cell cycle G2/M phase transition| |positive regulation of protein modification by small protein conjugation or removal| |regulation of protein dephosphorylation| |meiotic nuclear division| |meiotic cell cycle process| |peptidyl-serine phosphorylation| |regulation of phosphatase activity| |peptidyl-serine modification| |regulation of protein ubiquitination| |regulation of dephosphorylation| |meiotic cell cycle| |regulation of protein modification by small protein conjugation or removal| |mitotic cell cycle phase transition| |cell cycle phase transition| |nuclear division| |organelle fission| |cellular process involved in reproduction in multicellular organism| |protein serine/threonine kinase activity| |negative regulation of hydrolase activity| |centrosome| |cell division| |negative regulation of phosphate metabolic process| |negative regulation of phosphorus metabolic process| |negative regulation of protein modification process| |mitotic cell cycle process| |positive regulation of transferase activity| |mitotic cell cycle| |gamete generation| |cellular response to DNA damage stimulus| |negative regulation of catalytic activity| |multicellular organismal reproductive process| |sexual reproduction| |multicellular organism reproduction| |peptidyl-amino acid modification| |protein phosphorylation| |regulation of transferase activity| |multi-organism reproductive process| |cell cycle process| |negative regulation of cellular protein metabolic process| |negative regulation of protein metabolic process| |negative regulation of molecular function| |regulation of cell cycle| |positive regulation of protein modification process| |regulation of hydrolase activity| |phosphorylation| |cell cycle| |reproductive process| |reproduction| |positive regulation of catalytic activity| |ATP binding| |positive regulation of cellular protein metabolic process| |intracellular signal transduction| |cellular response to stress| |positive regulation of protein metabolic process| |positive regulation of molecular function| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of protein modification process| \\ === CRISPR Data === ^Screen^Score^ |BI-D1870 3.15μM R02 exp66|-2.19| |DABN 20μM R00 exp17|-2.1| |Nocodazole 0.2μM R02 exp64|-1.99| |Rotenone 2μM R00 exp33|-1.93| |BVD-523 15μM R02 exp67|-1.83| |Deferoxamine 11μM R07 exp350|-1.83| |Salinomycin 3.5μM R08 exp521|-1.77| |Oligomycin-A 2μM R00 exp25|-1.73| |Q15 2.7μM R02 exp79|-1.7| |VE-822 0.04μM R06 exp300|1.73| |KU-0063794 3.8μM R02 exp71|2.06| |SB203580 25μM R03 exp149|2.1| |A-196 10μM R03 exp114|2.12| ^Gene^Correlation^ |TSPY2|0.403| Global Fraction of Cell Lines Where Essential: 726/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|28/28| |blood|28/28| |bone|26/26| |breast|31/33| |central nervous system|56/56| |cervix|4/4| |colorectal|17/17| |esophagus|13/13| |fibroblast|1/1| |gastric|16/16| |kidney|21/21| |liver|19/20| |lung|75/75| |lymphocyte|16/16| |ovary|22/26| |pancreas|23/24| |peripheral nervous system|16/16| |plasma cell|15/15| |prostate|0/1| |skin|24/24| |soft tissue|9/9| |thyroid|2/2| |upper aerodigestive|22/22| |urinary tract|28/29| |uterus|4/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1217 * **Expression level (log2 read counts)**: 6.58
