This protein is preferentially expressed in cells of myeloid lineage. It interacts primarily with neutrophil cytosolic factor 2 (NCF2/p67-phox) to form a complex with neutrophil cytosolic factor 1 (NCF1/p47-phox), which further interacts with the small G protein RAC1 and translocates to the membrane upon cell stimulation. This complex then activates flavocytochrome b, the membrane-integrated catalytic core of the enzyme system. The PX domain of this protein can bind phospholipid products of the PI(3) kinase, which suggests its role in PI(3) kinase-mediated signaling events. The phosphorylation of this protein was found to negatively regulate the enzyme activity. Alternatively spliced transcript variants encoding distinct isoforms have been observed. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Component of the NADPH-oxidase, a multicomponent enzyme system responsible for the oxidative burst in which electrons are transported from NADPH to molecular oxygen, generating reactive oxidant intermediates. It may be important for the assembly and/or activation of the NADPH-oxidase complex. {ECO:0000269|PubMed:8280052}. |SH3 2| |PX| |SH3 1| |PB1| |phagolysosome| |superoxide-generating NADPH oxidase activator activity| |NADPH oxidase complex| |respiratory burst| |superoxide metabolic process| |phosphatidylinositol-3-phosphate binding| |vascular endothelial growth factor receptor signaling pathway| |antigen processing and presentation of exogenous peptide antigen via MHC class I, TAP-dependent| |cell redox homeostasis| |antigen processing and presentation of exogenous peptide antigen via MHC class I| |phosphatidylinositol binding| |antigen processing and presentation of peptide antigen via MHC class I| |reactive oxygen species metabolic process| |protein dimerization activity| |antigen processing and presentation of exogenous peptide antigen| |antigen processing and presentation of exogenous antigen| |antigen processing and presentation of peptide antigen| |endosome membrane| |antigen processing and presentation| |cellular response to oxidative stress| |phagocytosis| |response to oxidative stress| |transmembrane receptor protein tyrosine kinase signaling pathway| |enzyme linked receptor protein signaling pathway| |cellular homeostasis| |oxidation-reduction process| |positive regulation of catalytic activity| |homeostatic process| |cellular response to stress| |positive regulation of molecular function| |immune response| |vesicle-mediated transport| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp301|VER-155008 3.9μM R06 exp301]]|-1.94| |[[:results:exp60|Vinblastine 0.002μM R01 exp60]]|1.82| |[[:results:exp456|Benzoate 20000μM R08 exp456]]|1.91| |[[:results:exp100|NFN1 1μM R03 exp100]]|2.07| No correlation found to any other genes in chemogenomics.
