This protein is associated with the the histone deacetylase HDAC3 and with the Notch corepressor complex, and it thereby acts as a transcriptional repressor of Notch target genes. It is also required for alphabeta T cell development. A related pseudogene has been identified on chromosome X, while a related and intronless retrocopy, which has an intact CDS and may be functional, is located on chromosome 6. [provided by RefSeq, May 2010]. * **UniProt Summary**: Acts as a transcriptional repressor. Plays a role as a transcriptional corepressor of the Notch-mediated signaling required for T-cell development. Also involved in the TNF and IL-1 induced NF-kappa-B activation. Associates with chromatin at the Notch-regulated SKP2 promoter. {ECO:0000269|PubMed:14550261, ECO:0000269|PubMed:19409814}. |SynMuv product| |hematopoietic stem cell proliferation| |granulocyte differentiation| |positive regulation of alpha-beta T cell differentiation| |T cell differentiation in thymus| |stem cell proliferation| |positive regulation of alpha-beta T cell activation| |regulation of alpha-beta T cell differentiation| |chromatin DNA binding| |positive regulation of T cell differentiation| |regulation of alpha-beta T cell activation| |positive regulation of lymphocyte differentiation| |myeloid leukocyte differentiation| |Notch signaling pathway| |T cell differentiation| |stem cell population maintenance| |maintenance of cell number| |regulation of T cell differentiation| |positive regulation of leukocyte differentiation| |regulation of lymphocyte differentiation| |positive regulation of hemopoiesis| |positive regulation of T cell activation| |positive regulation of leukocyte cell-cell adhesion| |myeloid cell differentiation| |T cell activation| |lymphocyte differentiation| |positive regulation of cell-cell adhesion| |regulation of leukocyte differentiation| |regulation of leukocyte cell-cell adhesion| |regulation of T cell activation| |leukocyte differentiation| |positive regulation of lymphocyte activation| |lymphocyte activation| |chromatin binding| |regulation of cell-cell adhesion| |positive regulation of cell adhesion| |positive regulation of leukocyte activation| |positive regulation of cell activation| |regulation of hemopoiesis| |regulation of lymphocyte activation| |cell population proliferation| |hemopoiesis| |regulation of leukocyte activation| |hematopoietic or lymphoid organ development| |regulation of cell activation| |immune system development| |regulation of cell adhesion| |negative regulation of transcription by RNA polymerase II| |leukocyte activation| |positive regulation of cell differentiation| |cell activation| |positive regulation of immune system process| |negative regulation of transcription, DNA-templated| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |negative regulation of RNA metabolic process| |positive regulation of developmental process| |negative regulation of cellular macromolecule biosynthetic process| |RNA binding| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |regulation of immune system process| |negative regulation of gene expression| |positive regulation of multicellular organismal process| |regulation of cell differentiation| \\ === CRISPR Data === ^Screen^Score^ |AICAR 240μM R03 exp116|-2.33| |I-BRD9 4μM R07 exp365|-2.01| |Pimelic-diphenylamide-106 5μM R00 exp28|-2| |Dynasore 10μM R04 exp180|-1.92| |Losmapimod 1μM R07 exp376|-1.74| |Genistein 15μM R07 exp360|-1.73| |DABN racemic mixture R03 exp93|-1.7| |Rapamycin 2μM plus Cyclosporin-A 3μM R07 exp307|1.73| |JQ1 0.01μM R03 exp130|1.73| |NN-Diethyl-meta-toluamide 100μM R05 exp237|1.76| |Momordin-Ic 10μM R08 exp506|1.82| |Demecolcine 0.03μM R05 exp228|1.98| |UM0011500 5μM R05 exp245|2.07| |HMS-I1 1μM R01 exp46|2.61| ^Gene^Correlation^ |HGC6.3|0.441| |RAD51|0.406| Global Fraction of Cell Lines Where Essential: 606/694 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|24/28| |blood|24/26| |bone|25/26| |breast|23/30| |central nervous system|42/49| |cervix|3/4| |colorectal|16/17| |esophagus|9/11| |fibroblast|1/1| |gastric|13/14| |kidney|15/18| |liver|13/19| |lung|61/72| |lymphocyte|15/16| |ovary|22/25| |pancreas|17/22| |peripheral nervous system|13/15| |plasma cell|12/12| |prostate|1/1| |skin|19/20| |soft tissue|9/9| |thyroid|2/2| |upper aerodigestive|20/22| |urinary tract|26/28| |uterus|5/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1915 * **Expression level (log2 read counts)**: 5.26
