This complex catalyzes the overall conversion of 2-oxoglutarate (alpha-ketoglutarate) to succinyl-CoA and CO(2) during the Krebs cycle. The protein is located in the mitochondrial matrix and uses thiamine pyrophosphate as a cofactor. A congenital deficiency in 2-oxoglutarate dehydrogenase activity is believed to lead to hypotonia, metabolic acidosis, and hyperlactatemia. Alternative splicing results in multiple transcript variants encoding distinct isoforms.[provided by RefSeq, Sep 2009]. * **UniProt Summary**: 2-oxoglutarate dehydrogenase (E1) component of the 2- oxoglutarate dehydrogenase complex, which mediates the decarboxylation of alpha-ketoglutarate (PubMed:24495017). The 2- oxoglutarate dehydrogenase complex catalyzes the overall conversion of 2-oxoglutarate to succinyl-CoA and CO(2) (PubMed:24495017). The 2-oxoglutarate dehydrogenase complex is mainly active in the mitochondrion (PubMed:29211711). A fraction of the 2-oxoglutarate dehydrogenase complex also localizes in the nucleus and is required for lysine succinylation of histones: associates with KAT2A on chromatin and provides succinyl-CoA to histone succinyltransferase KAT2A (PubMed:29211711). {ECO:0000269|PubMed:24495017, ECO:0000269|PubMed:29211711}. |Transket pyr| |E1 dh| |oxoglutarate dehydrogenase (succinyl-transferring) activity| |protein succinylation| |histone succinylation| |thiamine pyrophosphate binding| |succinyl-CoA metabolic process| |oxoglutarate dehydrogenase complex| |lysine catabolic process| |lysine metabolic process| |2-oxoglutarate metabolic process| |aspartate family amino acid catabolic process| |tricarboxylic acid cycle| |glycolytic process| |ATP generation from ADP| |ADP metabolic process| |aspartate family amino acid metabolic process| |nucleoside diphosphate phosphorylation| |purine ribonucleoside diphosphate metabolic process| |nucleotide phosphorylation| |purine nucleoside diphosphate metabolic process| |ribonucleoside diphosphate metabolic process| |pyruvate metabolic process| |aerobic respiration| |nucleoside diphosphate metabolic process| |acyl-CoA metabolic process| |thioester metabolic process| |dicarboxylic acid metabolic process| |mitochondrial membrane| |alpha-amino acid catabolic process| |carbohydrate catabolic process| |cellular amino acid catabolic process| |ribonucleoside bisphosphate metabolic process| |purine nucleoside bisphosphate metabolic process| |nucleoside bisphosphate metabolic process| |cellular respiration| |protein acylation| |ATP metabolic process| |alpha-amino acid metabolic process| |energy derivation by oxidation of organic compounds| |coenzyme metabolic process| |carboxylic acid catabolic process| |organic acid catabolic process| |cellular amino acid metabolic process| |purine ribonucleotide metabolic process| |ribonucleotide metabolic process| |purine nucleotide metabolic process| |ribose phosphate metabolic process| |sulfur compound metabolic process| |purine-containing compound metabolic process| |mitochondrial matrix| |generation of precursor metabolites and energy| |small molecule catabolic process| |cofactor metabolic process| |nucleotide metabolic process| |nucleoside phosphate metabolic process| |carbohydrate metabolic process| |monocarboxylic acid metabolic process| |nucleobase-containing small molecule metabolic process| |cellular amide metabolic process| |organophosphate metabolic process| |carboxylic acid metabolic process| |oxidation-reduction process| |oxoacid metabolic process| |organic acid metabolic process| |carbohydrate derivative metabolic process| |organonitrogen compound catabolic process| |mitochondrion| |phosphorylation| |small molecule metabolic process| |organic substance catabolic process| |cellular catabolic process| \\ === CRISPR Data === ^Screen^Score^ |Bortezomib 0.05μM R00 exp7|-1.78| |Disulfiram 4.3μM R06 exp281|1.87| |Parthenolide 2.5μM R05 exp238|1.9| |Apcin 50 to 100μM on day4 R04 exp176|1.94| |A-196 10μM R03 exp114|2.11| |AICAR 240μM R05 exp211|2.68| ^Gene^Correlation^ |POLR2J3|0.476| |ADSS|0.427| |NARFL|0.413| |VPS52|0.409| |TBC1D3H|0.402| |SRGAP2C|0.402| Global Fraction of Cell Lines Where Essential: 55/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|1/28| |blood|7/28| |bone|0/25| |breast|7/33| |central nervous system|1/56| |cervix|1/4| |colorectal|1/17| |esophagus|1/13| |fibroblast|0/1| |gastric|0/15| |kidney|2/21| |liver|2/20| |lung|5/75| |lymphocyte|2/14| |ovary|1/26| |pancreas|1/24| |peripheral nervous system|0/16| |plasma cell|5/15| |prostate|0/1| |skin|2/24| |soft tissue|0/7| |thyroid|1/2| |upper aerodigestive|1/22| |urinary tract|3/29| |uterus|1/5| == Essentiality in NALM6 == * **Essentiality Rank**: 404 * **Expression level (log2 read counts)**: 7.01
