Most members contain an N-terminal pleckstrin homology domain and a highly conserved C-terminal OSBP-like sterol-binding domain, although some members contain only the sterol-binding domain. Transcript variants derived from alternative promoter usage and/or alternative splicing exist; they encode different isoforms. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Binds phospholipids; exhibits strong binding to phosphatidic acid and weak binding to phosphatidylinositol 3- phosphate (By similarity). Stabilizes GTP-bound RAB7A on late endosomes/lysosomes and alters functional properties of late endocytic compartments via its interaction with RAB7A (PubMed:16176980). Binds 25-hydroxycholesterol and cholesterol (PubMed:17428193). {ECO:0000250, ECO:0000269|PubMed:16176980, ECO:0000269|PubMed:17428193}. |Oxysterol BP| |Ank 2| |Ank| |sterol binding| |sterol transporter activity| |bile acid biosynthetic process| |bile acid metabolic process| |cholesterol binding| |sterol transport| |antigen processing and presentation of exogenous peptide antigen via MHC class II| |antigen processing and presentation of peptide or polysaccharide antigen via MHC class II| |antigen processing and presentation of peptide antigen via MHC class II| |phospholipid binding| |cholesterol metabolic process| |late endosome| |steroid biosynthetic process| |secondary alcohol metabolic process| |sterol metabolic process| |organic hydroxy compound transport| |lipid binding| |antigen processing and presentation of exogenous peptide antigen| |antigen processing and presentation of exogenous antigen| |monocarboxylic acid biosynthetic process| |organic hydroxy compound biosynthetic process| |antigen processing and presentation of peptide antigen| |antigen processing and presentation| |steroid metabolic process| |lipid transport| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |alcohol metabolic process| |lipid localization| |organic hydroxy compound metabolic process| |monocarboxylic acid metabolic process| |lipid biosynthetic process| |small molecule biosynthetic process| |intracellular membrane-bounded organelle| |carboxylic acid metabolic process| |oxoacid metabolic process| |organic acid metabolic process| |lipid metabolic process| |organic cyclic compound biosynthetic process| |small molecule metabolic process| |vesicle-mediated transport| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp126|GSK461364A 0.1μM R03 exp126]]|-1.8| |[[:results:exp7|Bortezomib 0.05μM R00 exp7]]|1.7| |[[:results:exp34|Rotenone 20μM R00 exp34]]|1.96| No correlation found to any other genes in chemogenomics.
