Pyruvate dehydrogenase kinases catalyze phosphorylation of serine residues of E1 to inactivate the E1 component and inhibit the complex. Pyruvate dehydrogenase phosphatases catalyze the dephosphorylation and activation of the E1 component to reverse the effects of pyruvate dehydrogenase kinases. Pyruvate dehydrogenase phosphatase is a heterodimer consisting of catalytic and regulatory subunits. Two catalytic subunits have been reported; one is predominantly expressed in skeletal muscle and another one is is much more abundant in the liver. The catalytic subunit, encoded by this gene, is the former, and belongs to the protein phosphatase 2C (PP2C) superfamily. Along with the pyruvate dehydrogenase complex and pyruvate dehydrogenase kinases, this enzyme is located in the mitochondrial matrix. Mutation in this gene causes pyruvate dehydrogenase phosphatase deficiency. Multiple alternatively spliced transcript variants encoding different isoforms have been identified.[provided by RefSeq, Jun 2009]. * **UniProt Summary**: Catalyzes the dephosphorylation and concomitant reactivation of the alpha subunit of the E1 component of the pyruvate dehydrogenase complex. {ECO:0000250}. |PP2C| |[pyruvate dehydrogenase (lipoamide)] phosphatase activity| |positive regulation of pyruvate dehydrogenase activity| |regulation of pyruvate dehydrogenase activity| |regulation of acetyl-CoA biosynthetic process from pyruvate| |regulation of acyl-CoA biosynthetic process| |regulation of coenzyme metabolic process| |regulation of sulfur metabolic process| |peptidyl-threonine dephosphorylation| |magnesium-dependent protein serine/threonine phosphatase activity| |regulation of cofactor metabolic process| |regulation of purine nucleotide biosynthetic process| |regulation of nucleotide biosynthetic process| |positive regulation of oxidoreductase activity| |protein serine/threonine phosphatase activity| |regulation of fatty acid metabolic process| |regulation of oxidoreductase activity| |regulation of purine nucleotide metabolic process| |regulation of nucleotide metabolic process| |regulation of cellular ketone metabolic process| |protein dephosphorylation| |dephosphorylation| |mitochondrial matrix| |regulation of lipid metabolic process| |regulation of cellular amide metabolic process| |regulation of small molecule metabolic process| |mitochondrion| |positive regulation of catalytic activity| |positive regulation of molecular function| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| \\ === CRISPR Data === ^Screen^Score^ |2-Deoxy-D-glucose 800μM R05 exp214|-1.87| |Nifuroxazide 5μM R03 exp102|3.1| ^Gene^Correlation^ |HGC6.3|0.587| |MPC1|0.463| |MPC2|0.433| |PDHB|0.419| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 10639 * **Expression level (log2 read counts)**: 4.39
