The protein is a calcium(2+)-independent cell-cell adhesion molecule that belongs to the immunoglobulin superfamily and has 3 extracellular immunoglobulin-like loops, a single transmembrane domain (in some isoforms), and a cytoplasmic region. This protein acts as a receptor for glycoprotein D (gD) of herpes simplex viruses 1 and 2 (HSV-1, HSV-2), and pseudorabies virus (PRV) and mediates viral entry into epithelial and neuronal cells. Mutations in this gene cause cleft lip and palate/ectodermal dysplasia 1 syndrome (CLPED1) as well as non-syndromic cleft lip with or without cleft palate (CL/P). Alternative splicing results in multiple transcript variants encoding proteins with distinct C-termini. [provided by RefSeq, Oct 2009]. * **UniProt Summary**: Promotes cell-cell contacts by forming homophilic or heterophilic trans-dimers. Heterophilic interactions have been detected between NECTIN1 and NECTIN3 and between NECTIN1 and NECTIN4. Has some neurite outgrowth-promoting activity. {ECO:0000269|PubMed:21980294}. |V-set| |ig| |C2-set 2| |growth cone membrane| |desmosome organization| |virion attachment to host cell| |virion binding| |adhesion of symbiont to host cell| |protein localization to cell junction| |enamel mineralization| |adhesion of symbiont to host| |tooth mineralization| |integral component of presynaptic active zone membrane| |apical junction complex| |cell-cell contact zone| |amelogenesis| |lens morphogenesis in camera-type eye| |hippocampal mossy fiber to CA3 synapse| |coreceptor activity| |heterophilic cell-cell adhesion via plasma membrane cell adhesion molecules| |cell adhesion molecule binding| |adherens junction| |iron ion transport| |virus receptor activity| |lens development in camera-type eye| |odontogenesis of dentin-containing tooth| |adherens junction organization| |biomineralization| |biomineral tissue development| |viral entry into host cell| |entry into host cell| |entry into host| |cell-cell adherens junction| |regulation of synapse assembly| |camera-type eye morphogenesis| |transition metal ion transport| |odontogenesis| |cell-cell junction organization| |retina development in camera-type eye| |eye morphogenesis| |interaction with host| |homophilic cell adhesion via plasma membrane adhesion molecules| |carbohydrate binding| |viral life cycle| |signaling receptor activity| |cell junction organization| |regulation of synapse organization| |regulation of synapse structure or activity| |sensory organ morphogenesis| |cell-cell adhesion via plasma-membrane adhesion molecules| |axon guidance| |neuron projection guidance| |camera-type eye development| |eye development| |visual system development| |sensory system development| |axonogenesis| |axon development| |dendrite| |cell morphogenesis involved in neuron differentiation| |neuron projection morphogenesis| |plasma membrane bounded cell projection morphogenesis| |protein heterodimerization activity| |cell projection morphogenesis| |cell-cell adhesion| |cell part morphogenesis| |sensory organ development| |chemotaxis| |taxis| |cell morphogenesis involved in differentiation| |metal ion transport| |neuron projection development| |intracellular membrane-bounded organelle| |viral process| |cell morphogenesis| |symbiotic process| |neuron development| |interspecies interaction between organisms| |cellular component morphogenesis| |cation transport| |protein homodimerization activity| |anatomical structure formation involved in morphogenesis| |regulation of nervous system development| |cell adhesion| |biological adhesion| |animal organ morphogenesis| |regulation of cellular component biogenesis| |neuron differentiation| |identical protein binding| |plasma membrane bounded cell projection organization| |cell projection organization| |locomotion| |ion transport| |generation of neurons| |movement of cell or subcellular component| |neurogenesis| |cell development| |tissue development| |immune response| |extracellular region| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp160|Ribavirin 10 to 15μM on day4 R04 exp160]]|-1.85| |[[:results:exp99|NFN1 0.4μM R03 exp99]]|-1.71| |[[:results:exp11|CCCP 1μM R00 exp11]]|-1.7| No correlation found to any other genes in chemogenomics.
