The gene product stimulates the GTPase activity of normal RAS p21 but not its oncogenic counterpart. Acting as a suppressor of RAS function, the protein enhances the weak intrinsic GTPase activity of RAS proteins resulting in the inactive GDP-bound form of RAS, thereby allowing control of cellular proliferation and differentiation. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2014]. * **UniProt Summary**: Inhibitory regulator of the Ras-cyclic AMP pathway. Binds inositol tetrakisphosphate (IP4). |C2| |RasGAP| |BTK| |PH| |negative regulation of Ras protein signal transduction| |negative regulation of small GTPase mediated signal transduction| |phospholipid binding| |regulation of Ras protein signal transduction| |GTPase activator activity| |regulation of small GTPase mediated signal transduction| |MAPK cascade| |signal transduction by protein phosphorylation| |positive regulation of GTPase activity| |regulation of GTPase activity| |negative regulation of intracellular signal transduction| |perinuclear region of cytoplasm| |positive regulation of hydrolase activity| |protein phosphorylation| |negative regulation of signal transduction| |regulation of hydrolase activity| |phosphorylation| |negative regulation of cell communication| |negative regulation of signaling| |positive regulation of catalytic activity| |negative regulation of response to stimulus| |intracellular signal transduction| |positive regulation of molecular function| |regulation of intracellular signal transduction| \\ === CRISPR Data === ^Screen^Score^ |Phorbol-12-myristate-13-acetate 0.57μM R08 exp514|-3.15| |Thapsigargin 0.005μM R07 exp407|1.76| |VER-155008 3.9μM R06 exp301|1.9| |I-BRD9 4μM R07 exp365|2.22| ^Gene^Correlation^ |NF1|0.457| |LZTR1|0.423| |DUSP6|0.421| |DGKD|0.406| Global Fraction of Cell Lines Where Essential: 2/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|1/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|1/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 17760 * **Expression level (log2 read counts)**: 5.01
