[provided by RefSeq, Jul 2008]. * **UniProt Summary**: E3 ubiquitin-protein ligase that mediates ubiquitination oF target proteins (PubMed:23277564, PubMed:24275455, PubMed:24981174). Depending on the associated E2 ligase, mediates 'Lys-48'- and 'Lys-63'-linked polyubiquitination of substrates (By similarity). Part of a BAG6-dependent quality control process ensuring that proteins of the secretory pathway that are mislocalized to the cytosol are degraded by the proteasome. Probably acts by providing the ubiquitin ligase activity associated with the BAG6 complex and be responsible for ubiquitination of the hydrophobic mislocalized proteins and their targeting to the proteasome (PubMed:24981174). May also play a role in the endosomal recycling of IGF2R, the cation-independent mannose-6-phosphate receptor (PubMed:24275455). May play a role in the endosomal sorting and degradation of several membrane receptors including EGFR, FLT3, MET and CXCR4, by mediating their ubiquitination (PubMed:23418353). By ubiquitinating CDKN1A/p21 and targeting it for degradation, may also promote cell proliferation (PubMed:23026136). May monoubiquitinate AICDA (PubMed:23277564). {ECO:0000250|UniProtKB:Q91YL2, ECO:0000269|PubMed:23277564, ECO:0000269|PubMed:23418353, ECO:0000269|PubMed:24275455, ECO:0000269|PubMed:24981174, ECO:0000305|PubMed:23026136}. |zf-C3HC4| |zf-rbx1| |cytoplasm protein quality control by the ubiquitin-proteasome system| |ubiquitin-dependent protein catabolic process via the multivesicular body sorting pathway| |cellular response to misfolded protein| |response to misfolded protein| |protein quality control for misfolded or incompletely synthesized proteins| |epidermal growth factor receptor binding| |protein K63-linked ubiquitination| |negative regulation of epidermal growth factor receptor signaling pathway| |negative regulation of ERBB signaling pathway| |protein K48-linked ubiquitination| |protein monoubiquitination| |regulation of epidermal growth factor receptor signaling pathway| |regulation of ERBB signaling pathway| |retrograde transport, endosome to Golgi| |cytosolic transport| |cellular response to topologically incorrect protein| |response to topologically incorrect protein| |ubiquitin protein ligase activity| |endosomal transport| |protein polyubiquitination| |proteasome-mediated ubiquitin-dependent protein catabolic process| |proteasomal protein catabolic process| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |protein catabolic process| |protein ubiquitination| |protein modification by small protein conjugation| |cellular macromolecule catabolic process| |protein modification by small protein conjugation or removal| |macromolecule catabolic process| |organonitrogen compound catabolic process| |negative regulation of signal transduction| |proteolysis| |negative regulation of cell communication| |negative regulation of signaling| |intracellular transport| |regulation of cell population proliferation| |negative regulation of response to stimulus| |cellular response to stress| |organic substance catabolic process| |cellular catabolic process| |establishment of localization in cell| |vesicle-mediated transport| \\ === CRISPR Data === ^Screen^Score^ |Ethanol 0.01 R05 exp234|-1.81| |Dynasore 10μM R04 exp180|-1.75| |Benzoate 20000μM R08 exp456|1.71| |PU-H71 1μM R08 exp515|1.79| |42°C R08 exp539|1.91| |CAY10603 0.55μM R08 exp467|1.91| |HMS-I1 1μM R01 exp46|1.95| |UMK57 1μM R01 exp59|1.98| ^Gene^Correlation^ |KCMF1|0.502| |RCE1|0.426| |OTUD5|0.419| |AARS|0.41| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 2766 * **Expression level (log2 read counts)**: 5.67
