The protein encoded by this gene cross-links to a region in the pre-mRNA immediately upstream of the branchpoint sequence in pre-mRNA in the prespliceosomal complex A. It also may be involved in the assembly of the B, C and E spliceosomal complexes. In addition to RNA-binding activity, this protein interacts directly and highly specifically with subunit 2 of the splicing factor 3B. This protein contains two N-terminal RNA-recognition motifs (RRMs), consistent with the observation that it binds directly to pre-mRNA. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Involved in pre-mRNA splicing as a component of the splicing factor SF3B complex (PubMed:27720643). SF3B complex is required for 'A' complex assembly formed by the stable binding of U2 snRNP to the branchpoint sequence (BPS) in pre-mRNA. Sequence independent binding of SF3A/SF3B complex upstream of the branch site is essential, it may anchor U2 snRNP to the pre-mRNA (PubMed:12234937). May also be involved in the assembly of the 'E' complex. SF3B4 has been found in complex 'B' and 'C' as well (PubMed:10882114). Belongs also to the minor U12-dependent spliceosome, which is involved in the splicing of rare class of nuclear pre-mRNA intron (PubMed:15146077). {ECO:0000269|PubMed:10882114, ECO:0000269|PubMed:12234937, ECO:0000269|PubMed:15146077, ECO:0000269|PubMed:27720643}. |RRM 1| |positive regulation of mRNA splicing, via spliceosome| |U12-type spliceosomal complex| |positive regulation of mRNA processing| |positive regulation of RNA splicing| |U2-type precatalytic spliceosome| |positive regulation of mRNA metabolic process| |spliceosomal complex| |regulation of mRNA splicing, via spliceosome| |regulation of RNA splicing| |regulation of mRNA processing| |ribonucleoprotein complex| |RNA splicing, via transesterification reactions with bulged adenosine as nucleophile| |mRNA splicing, via spliceosome| |RNA splicing, via transesterification reactions| |regulation of mRNA metabolic process| |RNA splicing| |mRNA processing| |mRNA metabolic process| |nucleolus| |RNA processing| |RNA binding| |RNA metabolic process| |positive regulation of RNA metabolic process| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of gene expression| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Actinomycin-D 0.001μM R00 exp3|-1.74| |Lead acetate 2000μM R08 exp497|1.8| |Prochlorperazine 5.2μM R02 exp77|1.85| |THZ531 0.225μM R08 exp444|1.89| |Cycloheximide 0.2μM R00 exp15|1.96| |Lead acetate 2000μM R08 exp498 no dilution day6|2.16| ^Gene^Correlation^ |TERF1|0.413| |PSMB4|0.404| Global Fraction of Cell Lines Where Essential: 739/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|28/28| |blood|28/28| |bone|26/26| |breast|33/33| |central nervous system|56/56| |cervix|4/4| |colorectal|17/17| |esophagus|13/13| |fibroblast|1/1| |gastric|16/16| |kidney|21/21| |liver|20/20| |lung|75/75| |lymphocyte|16/16| |ovary|26/26| |pancreas|24/24| |peripheral nervous system|16/16| |plasma cell|15/15| |prostate|1/1| |skin|24/24| |soft tissue|9/9| |thyroid|2/2| |upper aerodigestive|22/22| |urinary tract|29/29| |uterus|5/5| == Essentiality in NALM6 == * **Essentiality Rank**: 408 * **Expression level (log2 read counts)**: 6.29
