The encoded protein, either alone or when in the SWI/SNF complex, can bind to 4-way junction DNA, which is thought to mimic the topology of DNA as it enters or exits the nucleosome. The protein contains a DNA-binding HMG domain, but disruption of this domain does not abolish the DNA-binding or nucleosome-displacement activities of the SWI/SNF complex. Unlike most of the SWI/SNF complex proteins, this protein has no yeast counterpart. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Involved in transcriptional activation and repression of select genes by chromatin remodeling (alteration of DNA-nucleosome topology). Component of SWI/SNF chromatin remodeling complexes that carry out key enzymatic activities, changing chromatin structure by altering DNA-histone contacts within a nucleosome in an ATP-dependent manner. Belongs to the neural progenitors- specific chromatin remodeling complex (npBAF complex) and the neuron-specific chromatin remodeling complex (nBAF complex). During neural development a switch from a stem/progenitor to a postmitotic chromatin remodeling mechanism occurs as neurons exit the cell cycle and become committed to their adult state. The transition from proliferating neural stem/progenitor cells to postmitotic neurons requires a switch in subunit composition of the npBAF and nBAF complexes. As neural progenitors exit mitosis and differentiate into neurons, npBAF complexes which contain ACTL6A/BAF53A and PHF10/BAF45A, are exchanged for homologous alternative ACTL6B/BAF53B and DPF1/BAF45B or DPF3/BAF45C subunits in neuron-specific complexes (nBAF). The npBAF complex is essential for the self-renewal/proliferative capacity of the multipotent neural stem cells. The nBAF complex along with CREST plays a role regulating the activity of genes essential for dendrite growth (By similarity). Required for the coactivation of estrogen responsive promoters by SWI/SNF complexes and the SRC/p160 family of histone acetyltransferases (HATs). Also specifically interacts with the CoREST corepressor resulting in repression of neuronal specific gene promoters in non-neuronal cells. {ECO:0000250|UniProtKB:O54941, ECO:0000303|PubMed:12672490, ECO:0000303|PubMed:22952240, ECO:0000303|PubMed:26601204}. |HMG box| |N-acetyltransferase activity| |npBAF complex| |nBAF complex| |SWI/SNF complex| |nucleosome disassembly| |chromatin disassembly| |nuclear receptor binding| |protein-DNA complex disassembly| |nuclear chromosome| |nucleosomal DNA binding| |ATP-dependent chromatin remodeling| |RNA polymerase II distal enhancer sequence-specific DNA binding| |protein N-terminus binding| |chromatin assembly or disassembly| |nucleosome organization| |chromatin remodeling| |nuclear chromatin| |protein-containing complex disassembly| |protein-DNA complex subunit organization| |transcription coactivator activity| |chromatin binding| |cellular component disassembly| |RNA polymerase II proximal promoter sequence-specific DNA binding| |protein-containing complex| |chromatin organization| |chromosome organization| |negative regulation of transcription, DNA-templated| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |negative regulation of RNA metabolic process| |negative regulation of cellular macromolecule biosynthetic process| |RNA binding| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |DNA-binding transcription factor activity, RNA polymerase II-specific| |neurogenesis| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |negative regulation of gene expression| |positive regulation of RNA metabolic process| |protein-containing complex subunit organization| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Momordin-Ic 10μM R08 exp506|1.71| |NFN1 0.1μM R01 exp49|1.73| |TNF-alpha 44ng/ml R08 exp533|1.92| |BI-6727 0.001μM R01 exp42|2.32| |TRAIL 50ng/ml R00 exp36|3.52| |TRAIL 5ng/ml R00 exp35|3.54| ^Gene^Correlation^ |CXorf38|0.441| |CASP8|0.407| |C2CD2L|0.403| Global Fraction of Cell Lines Where Essential: 68/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|6/28| |bone|0/25| |breast|10/33| |central nervous system|2/56| |cervix|1/4| |colorectal|1/17| |esophagus|1/13| |fibroblast|0/1| |gastric|2/15| |kidney|1/21| |liver|0/20| |lung|5/75| |lymphocyte|3/14| |ovary|4/26| |pancreas|2/24| |peripheral nervous system|1/16| |plasma cell|2/15| |prostate|1/1| |skin|2/24| |soft tissue|0/7| |thyroid|0/2| |upper aerodigestive|3/22| |urinary tract|2/29| |uterus|1/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1765 * **Expression level (log2 read counts)**: 7.2
