The main functional protein encoded by this gene is a humoral growth factor that is necessary for megakaryocyte proliferation and maturation, as well as for thrombopoiesis. This protein is the ligand for MLP/C_MPL, the product of myeloproliferative leukemia virus oncogene. Mutations in this gene are the cause of thrombocythemia 1. Alternative promoter usage and differential splicing result in multiple transcript variants differing in the 5' UTR and/or coding region. Multiple AUG codons upstream of the main open reading frame (ORF) have been identified, and these upstream AUGs inhibit translation of the main ORF at different extent. [provided by RefSeq, Feb 2014]. * **UniProt Summary**: Lineage-specific cytokine affecting the proliferation and maturation of megakaryocytes from their committed progenitor cells. It acts at a late stage of megakaryocyte development. It may be the major physiological regulator of circulating platelets. |EPO TPO| |positive regulation of hematopoietic stem cell proliferation| |thrombopoietin-mediated signaling pathway| |positive regulation of megakaryocyte differentiation| |regulation of hematopoietic stem cell proliferation| |megakaryocyte development| |megakaryocyte differentiation| |bone cell development| |positive regulation of stem cell proliferation| |receptor signaling pathway via STAT| |regulation of megakaryocyte differentiation| |myeloid cell development| |regulation of stem cell proliferation| |chemokine-mediated signaling pathway| |response to chemokine| |cellular response to chemokine| |positive regulation of myeloid cell differentiation| |hormone activity| |growth factor activity| |positive regulation of protein kinase B signaling| |cytokine activity| |positive regulation of hemopoiesis| |bone development| |positive regulation of ERK1 and ERK2 cascade| |regulation of protein kinase B signaling| |myeloid cell differentiation| |regulation of myeloid cell differentiation| |regulation of ERK1 and ERK2 cascade| |signaling receptor binding| |regulation of hemopoiesis| |skeletal system development| |positive regulation of MAPK cascade| |cell population proliferation| |hemopoiesis| |hematopoietic or lymphoid organ development| |immune system development| |cytokine-mediated signaling pathway| |regulation of MAPK cascade| |positive regulation of cell population proliferation| |positive regulation of cell differentiation| |cellular response to cytokine stimulus| |positive regulation of protein phosphorylation| |positive regulation of intracellular signal transduction| |positive regulation of phosphorylation| |response to cytokine| |positive regulation of phosphorus metabolic process| |positive regulation of phosphate metabolic process| |positive regulation of immune system process| |positive regulation of protein modification process| |positive regulation of developmental process| |regulation of protein phosphorylation| |regulation of phosphorylation| |extracellular space| |positive regulation of cellular protein metabolic process| |regulation of cell population proliferation| |cell development| |positive regulation of signal transduction| |regulation of immune system process| |positive regulation of protein metabolic process| |positive regulation of multicellular organismal process| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |regulation of cell differentiation| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |regulation of protein modification process| |extracellular region| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp536|Vitamin-D3 40μM R08 exp536]]|-2.1| |[[:results:exp25|Oligomycin-A 2μM R00 exp25]]|-1.94| |[[:results:exp439|QNZ 0.01μM R08 exp439]]|-1.92| |[[:results:exp160|Ribavirin 10 to 15μM on day4 R04 exp160]]|-1.87| No correlation found to any other genes in chemogenomics.
