This receptor is activated by tumor necrosis factor-related apoptosis inducing ligand (TNFSF10/TRAIL), and thus transduces cell death signal and induces cell apoptosis. Studies with FADD-deficient mice suggested that FADD, a death domain containing adaptor protein, is required for the apoptosis mediated by this protein. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Receptor for the cytotoxic ligand TNFSF10/TRAIL (PubMed:26457518). The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. Promotes the activation of NF-kappa-B. {ECO:0000269|PubMed:26457518, ECO:0000269|PubMed:9430227}. |Death| |TNFR c6| |death receptor activity| |TRAIL binding| |TRAIL-activated apoptotic signaling pathway| |activation of NF-kappaB-inducing kinase activity| |extrinsic apoptotic signaling pathway via death domain receptors| |negative regulation of extrinsic apoptotic signaling pathway via death domain receptors| |regulation of extrinsic apoptotic signaling pathway via death domain receptors| |positive regulation of NIK/NF-kappaB signaling| |cellular response to mechanical stimulus| |NIK/NF-kappaB signaling| |activation of cysteine-type endopeptidase activity involved in apoptotic process| |extrinsic apoptotic signaling pathway| |negative regulation of extrinsic apoptotic signaling pathway| |protease binding| |regulation of NIK/NF-kappaB signaling| |positive regulation of cysteine-type endopeptidase activity involved in apoptotic process| |positive regulation of cysteine-type endopeptidase activity| |regulation of extrinsic apoptotic signaling pathway| |positive regulation of endopeptidase activity| |positive regulation of peptidase activity| |signaling receptor activity| |response to mechanical stimulus| |regulation of cysteine-type endopeptidase activity involved in apoptotic process| |negative regulation of apoptotic signaling pathway| |regulation of cysteine-type endopeptidase activity| |apoptotic signaling pathway| |cellular response to abiotic stimulus| |cellular response to environmental stimulus| |activation of protein kinase activity| |transcription factor binding| |cellular response to external stimulus| |positive regulation of proteolysis| |leukocyte migration| |regulation of apoptotic signaling pathway| |regulation of endopeptidase activity| |regulation of peptidase activity| |positive regulation of protein kinase activity| |positive regulation of kinase activity| |cell surface| |positive regulation of apoptotic process| |positive regulation of programmed cell death| |positive regulation of transferase activity| |positive regulation of cell death| |regulation of proteolysis| |positive regulation of hydrolase activity| |regulation of protein kinase activity| |regulation of kinase activity| |negative regulation of apoptotic process| |negative regulation of programmed cell death| |apoptotic process| |cell migration| |regulation of transferase activity| |negative regulation of cell death| |positive regulation of protein phosphorylation| |positive regulation of intracellular signal transduction| |programmed cell death| |positive regulation of phosphorylation| |localization of cell| |cell motility| |cell death| |positive regulation of phosphate metabolic process| |positive regulation of phosphorus metabolic process| |response to abiotic stimulus| |positive regulation of protein modification process| |negative regulation of signal transduction| |regulation of hydrolase activity| |locomotion| |negative regulation of cell communication| |negative regulation of signaling| |positive regulation of catalytic activity| |regulation of protein phosphorylation| |regulation of apoptotic process| |movement of cell or subcellular component| |regulation of programmed cell death| |regulation of phosphorylation| |positive regulation of cellular protein metabolic process| |negative regulation of response to stimulus| |positive regulation of signal transduction| |regulation of cell death| |intracellular signal transduction| |positive regulation of protein metabolic process| |positive regulation of molecular function| |regulation of phosphate metabolic process| |regulation of phosphorus metabolic process| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |regulation of protein modification process| \\ === CRISPR Data === ^Screen^Score^ |NN-Diethyl-meta-toluamide 100μM R05 exp237|-1.86| ^Gene^Correlation^ |POLR2J3|0.461| |HGC6.3|0.444| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 11735 * **Expression level (log2 read counts)**: 1.49
