This cytokine is a ligand for receptors TNFRSF13B/TACI, TNFRSF17/BCMA, and TNFRSF13C/BAFFR. This cytokine is expressed in B cell lineage cells, and acts as a potent B cell activator. It has been also shown to play an important role in the proliferation and differentiation of B cells. Alternatively spliced transcript variants encoding distinct isoforms have been identified. [provided by RefSeq, Mar 2011]. * **UniProt Summary**: Cytokine that binds to TNFRSF13B/TACI and TNFRSF17/BCMA. TNFSF13/APRIL binds to the same 2 receptors. Together, they form a 2 ligands -2 receptors pathway involved in the stimulation of B- and T-cell function and the regulation of humoral immunity. A third B-cell specific BAFF-receptor (BAFFR/BR3) promotes the survival of mature B-cells and the B-cell response. {ECO:0000269|PubMed:10973284}. Isoform 3: Acts as a transcription factor for its own parent gene, in association with NF-kappa-B p50 subunit, at least in autoimmune and proliferative B-cell diseases. The presence of Delta4BAFF is essential for soluble BAFF release by IFNG/IFN- gamma-stimulated monocytes and for B-cell survival. It can directly or indirectly regulate the differential expression of a large number of genes involved in the innate immune response and the regulation of apoptosis. {ECO:0000269|PubMed:10973284}. |TNF| |positive regulation of germinal center formation| |B cell costimulation| |immunoglobulin secretion| |regulation of germinal center formation| |B cell homeostasis| |tumor necrosis factor receptor binding| |positive regulation of B cell proliferation| |lymphocyte homeostasis| |T cell costimulation| |lymphocyte costimulation| |regulation of B cell proliferation| |leukocyte homeostasis| |positive regulation of T cell proliferation| |positive regulation of adaptive immune response based on somatic recombination of immune receptors built from immunoglobulin superfamily domains| |positive regulation of adaptive immune response| |tumor necrosis factor-mediated signaling pathway| |positive regulation of lymphocyte proliferation| |positive regulation of mononuclear cell proliferation| |positive regulation of leukocyte proliferation| |regulation of adaptive immune response based on somatic recombination of immune receptors built from immunoglobulin superfamily domains| |regulation of T cell proliferation| |immunoglobulin production| |protein secretion| |establishment of protein localization to extracellular region| |regulation of adaptive immune response| |protein localization to extracellular region| |positive regulation of B cell activation| |peptide secretion| |cytokine activity| |production of molecular mediator of immune response| |homeostasis of number of cells| |positive regulation of T cell activation| |regulation of B cell activation| |regulation of lymphocyte proliferation| |regulation of mononuclear cell proliferation| |positive regulation of leukocyte cell-cell adhesion| |regulation of leukocyte proliferation| |cellular response to tumor necrosis factor| |positive regulation of cell-cell adhesion| |response to tumor necrosis factor| |regulation of leukocyte cell-cell adhesion| |regulation of T cell activation| |signaling receptor binding| |positive regulation of lymphocyte activation| |regulation of cell-cell adhesion| |positive regulation of cell adhesion| |positive regulation of leukocyte activation| |positive regulation of cell activation| |regulation of lymphocyte activation| |regulation of leukocyte activation| |regulation of cell activation| |cytokine-mediated signaling pathway| |regulation of cell adhesion| |perinuclear region of cytoplasm| |positive regulation of immune response| |positive regulation of cell population proliferation| |secretion by cell| |cellular response to cytokine stimulus| |export from cell| |regulation of anatomical structure morphogenesis| |response to cytokine| |secretion| |positive regulation of immune system process| |regulation of immune response| |positive regulation of developmental process| |protein transport| |peptide transport| |amide transport| |extracellular space| |establishment of protein localization| |regulation of cell population proliferation| |homeostatic process| |regulation of immune system process| |nitrogen compound transport| |immune response| |extracellular region| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp474|CR131-b 0.005μM R08 exp474]]|1.75| |[[:results:exp335|Aminopterin 0.005μM R07 exp335]]|1.83| No correlation found to any other genes in chemogenomics.
