The TRIM motif includes three zinc-binding domains, a RING, a B-box type 1 and a B-box type 2, and a coiled-coil region. The protein localizes to cytoplasmic filaments. It plays a neuroprotective role and functions as an E3-ubiquitin ligase in proteasome-mediated degradation of target proteins. Mutations in this gene can cause early-onset axonal neuropathy. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Nov 2014]. * **UniProt Summary**: UBE2D1-dependent E3 ubiquitin-protein ligase that mediates the ubiquitination of NEFL and of phosphorylated BCL2L11. Plays a neuroprotective function. May play a role in neuronal rapid ischemic tolerance. {ECO:0000250|UniProtKB:Q9ESN6}. |NHL| |Filamin| |zf-C3HC4| |zf-B box| |regulation of neuron apoptotic process| |ubiquitin protein ligase activity| |ubiquitin-protein transferase activity| |protein polyubiquitination| |regulation of neuron death| |proteasome-mediated ubiquitin-dependent protein catabolic process| |proteasomal protein catabolic process| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |modification-dependent macromolecule catabolic process| |proteolysis involved in cellular protein catabolic process| |cellular protein catabolic process| |protein catabolic process| |protein ubiquitination| |protein modification by small protein conjugation| |zinc ion binding| |cellular macromolecule catabolic process| |protein modification by small protein conjugation or removal| |macromolecule catabolic process| |organonitrogen compound catabolic process| |proteolysis| |regulation of apoptotic process| |regulation of programmed cell death| |regulation of cell death| |organic substance catabolic process| |cellular catabolic process| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp470|Chloroquine 32μM R08 exp470]]|1.71| ^Gene^Correlation^ |[[:human genes:p:polr2j3|POLR2J3]]|0.4| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 9579 * **Expression level (log2 read counts)**: 3.53 {{:chemogenomics:nalm6 dist.png?nolink |}}
