This endonuclease catalyzes the first step in RNA splicing which is the removal of introns. Mutations in this gene have been associated with pontocerebellar hypoplasia type 2. A pseudogene has been identified on chromosome 4. Multiple transcript variants encoding different isoforms have been found for this gene.[provided by RefSeq, Feb 2009]. * **UniProt Summary**: Constitutes one of the two catalytic subunit of the tRNA-splicing endonuclease complex, a complex responsible for identification and cleavage of the splice sites in pre-tRNA. It cleaves pre-tRNA at the 5'- and 3'-splice sites to release the intron. The products are an intron and two tRNA half-molecules bearing 2',3'-cyclic phosphate and 5'-OH termini. There are no conserved sequences at the splice sites, but the intron is invariably located at the same site in the gene, placing the splice sites an invariant distance from the constant structural features of the tRNA body. Isoform 1 probably carries the active site for 5'-splice site cleavage. The tRNA splicing endonuclease is also involved in mRNA processing via its association with pre- mRNA 3'-end processing factors, establishing a link between pre- tRNA splicing and pre-mRNA 3'-end formation, suggesting that the endonuclease subunits function in multiple RNA-processing events. Isoform 2 is responsible for processing a yet unknown RNA substrate. The complex containing isoform 2 is not able to cleave pre-tRNAs properly, although it retains endonucleolytic activity. {ECO:0000269|PubMed:15109492}. |tRNA int endo| |tRNA int endo N| |tRNA-type intron splice site recognition and cleavage| |tRNA-intron endonuclease activity| |tRNA-intron endonuclease complex| |lyase activity| |tRNA splicing, via endonucleolytic cleavage and ligation| |RNA splicing, via endonucleolytic cleavage and ligation| |RNA phosphodiester bond hydrolysis, endonucleolytic| |tRNA processing| |RNA phosphodiester bond hydrolysis| |tRNA metabolic process| |nucleic acid binding| |nucleic acid phosphodiester bond hydrolysis| |ncRNA processing| |RNA splicing| |ncRNA metabolic process| |mRNA processing| |centrosome| |mRNA metabolic process| |nucleolus| |RNA processing| |RNA metabolic process| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Benzoate 20000μM R08 exp456|1.71| |∆-9-Tetrahydrocannabinol 30μM R08 exp445|1.71| |Nicotinate 1000μM R03 exp140|1.75| |Etoposide 1μM R00 exp19|1.77| |MLN-4924 2μM R00 exp22|1.96| |Pimelic-diphenylamide-106 5μM R00 exp28|2.27| ^Gene^Correlation^ |HARS|0.569| |EPRS|0.494| |GNB1L|0.448| |RPTOR|0.445| |AARS|0.444| |RTCB|0.443| |EIF4E|0.441| |GMPPB|0.44| |POLR3H|0.436| |ZNF259|0.436| |VRK1|0.435| |POLR3B|0.432| |MTOR|0.431| |TTI2|0.42| |GTF3C3|0.418| |TSEN34|0.418| |NAE1|0.417| |PAM16|0.415| |PMPCB|0.413| |GFPT1|0.413| |GTF3A|0.413| |ALG13|0.408| |HSPD1|0.406| |UAP1|0.402| |SRP68|0.401| Global Fraction of Cell Lines Where Essential: 145/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|6/28| |blood|2/28| |bone|6/26| |breast|1/33| |central nervous system|6/56| |cervix|1/4| |colorectal|3/17| |esophagus|3/13| |fibroblast|0/1| |gastric|1/16| |kidney|6/21| |liver|6/20| |lung|14/75| |lymphocyte|1/16| |ovary|5/26| |pancreas|9/24| |peripheral nervous system|4/16| |plasma cell|0/15| |prostate|0/1| |skin|10/24| |soft tissue|1/9| |thyroid|0/2| |upper aerodigestive|6/22| |urinary tract|2/29| |uterus|2/5| == Essentiality in NALM6 == * **Essentiality Rank**: 1551 * **Expression level (log2 read counts)**: 5.18
