Recognizes and binds to proteins bearing specific N-terminal residues that are destabilizing according to the N-end rule, leading to their ubiquitination and subsequent degradation. Plays a critical role in chromatin inactivation and chromosome-wide transcriptional silencing during meiosis via ubiquitination of histone H2A. Binds leucine and is a negative regulator of the leucine-mTOR signaling pathway, thereby controlling cell growth. Required for spermatogenesis, promotes, with Tex19.1, SPO11-dependent recombination foci to accumulate and drive robust homologous chromosome synapsis (By similarity). Polyubiquitinates LINE-1 retrotransposon encoded, LIRE1, which induces degradation, inhibiting LINE-1 retranstoposon mobilization (By similarity). {ECO:0000250|UniProtKB:Q6WKZ8, ECO:0000269|PubMed:15548684, ECO:0000269|PubMed:20298436, ECO:0000269|PubMed:20835242}. |ClpS| |zf-UBR| |ubiquitin-dependent protein catabolic process via the N-end rule pathway| |leucine binding| |cellular response to leucine| |response to leucine| |negative regulation of transposition| |regulation of transposition| |histone H2A ubiquitination| |male meiosis I| |histone ubiquitination| |male meiotic nuclear division| |negative regulation of TOR signaling| |reciprocal meiotic recombination| |homologous recombination| |chromatin silencing| |chromatin organization involved in negative regulation of transcription| |cellular response to amino acid stimulus| |chromatin organization involved in regulation of transcription| |negative regulation of gene expression, epigenetic| |regulation of TOR signaling| |ubiquitin ligase complex| |meiosis I| |meiosis I cell cycle process| |response to amino acid| |chromatin| |meiotic nuclear division| |gene silencing| |meiotic cell cycle process| |cellular response to acid chemical| |ubiquitin protein ligase activity| |DNA recombination| |meiotic cell cycle| |regulation of gene expression, epigenetic| |nuclear division| |protein polyubiquitination| |organelle fission| |proteasome-mediated ubiquitin-dependent protein catabolic process| |response to acid chemical| |proteasomal protein catabolic process| |cellular process involved in reproduction in multicellular organism| |histone modification| |covalent chromatin modification| |negative regulation of intracellular signal transduction| |ubiquitin-dependent protein catabolic process| |modification-dependent protein catabolic process| |spermatogenesis| |modification-dependent macromolecule catabolic process| |male gamete generation| |proteolysis involved in cellular protein catabolic process| |cellular response to organonitrogen compound| |cellular protein catabolic process| |cellular response to nitrogen compound| |protein catabolic process| |protein ubiquitination| |gamete generation| |chromatin organization| |DNA metabolic process| |protein modification by small protein conjugation| |multicellular organismal reproductive process| |sexual reproduction| |zinc ion binding| |multicellular organism reproduction| |cellular macromolecule catabolic process| |protein modification by small protein conjugation or removal| |multi-organism reproductive process| |cell cycle process| |response to organonitrogen compound| |macromolecule catabolic process| |cellular response to oxygen-containing compound| |organonitrogen compound catabolic process| |chromosome organization| |response to nitrogen compound| |negative regulation of transcription, DNA-templated| |cellular response to endogenous stimulus| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |negative regulation of signal transduction| |proteolysis| |negative regulation of RNA metabolic process| |cell cycle| |negative regulation of cell communication| |negative regulation of signaling| |negative regulation of cellular macromolecule biosynthetic process| |reproductive process| |reproduction| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |response to endogenous stimulus| |negative regulation of cellular biosynthetic process| |negative regulation of biosynthetic process| |response to oxygen-containing compound| |negative regulation of response to stimulus| |negative regulation of gene expression| |organic substance catabolic process| |cellular catabolic process| |regulation of intracellular signal transduction| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp450|Artemisinin 50μM R08 exp450]]|-1.78| |[[:results:exp295|Pyronaridine 1μM R06 exp295]]|1.72| |[[:results:exp416|Tubacin 1.6μM R07 exp416]]|1.74| |[[:results:exp234|Ethanol 0.01 R05 exp234]]|1.84| |[[:results:exp6|Bortezomib 0.005μM R00 exp6]]|2.84| |[[:results:exp468|CB-5083 0.4μM R08 exp468]]|2.92| No correlation found to any other genes in chemogenomics.
