This gene encodes a protein that functions as a specific guanine nucleotide exchange factor for RhoA. It also interacts with ephrin A4 in vascular smooth muscle cells. Two alternatively spliced transcripts variants that encode the same protein have been found for this gene. [provided by RefSeq, Aug 2010]. * **UniProt Summary**: Specific GEF for RhoA activation. Does not activate RAC1 or CDC42. Regulates vascular smooth muscle contractility. Negatively regulates excitatory synapse development by suppressing the synapse-promoting activity of EPHB2. {ECO:0000269|PubMed:12775584}. |RhoGEF| |negative regulation of synapse maturation| |retina vasculature morphogenesis in camera-type eye| |regulation of synapse maturation| |retina vasculature development in camera-type eye| |negative regulation of synapse organization| |positive regulation of stress fiber assembly| |positive regulation of actin filament bundle assembly| |Rho guanyl-nucleotide exchange factor activity| |regulation of stress fiber assembly| |regulation of actomyosin structure organization| |regulation of actin filament bundle assembly| |retina development in camera-type eye| |regulation of Rho protein signal transduction| |positive regulation of supramolecular fiber organization| |regulation of synapse organization| |positive regulation of cytoskeleton organization| |regulation of synapse structure or activity| |regulation of Ras protein signal transduction| |regulation of actin filament organization| |GTPase activator activity| |negative regulation of nervous system development| |camera-type eye development| |regulation of actin cytoskeleton organization| |regulation of small GTPase mediated signal transduction| |regulation of supramolecular fiber organization| |eye development| |visual system development| |sensory system development| |regulation of actin filament-based process| |positive regulation of GTPase activity| |dendrite| |regulation of GTPase activity| |vasculature development| |cardiovascular system development| |positive regulation of cellular component biogenesis| |regulation of cytoskeleton organization| |sensory organ development| |positive regulation of organelle organization| |negative regulation of cellular component organization| |positive regulation of hydrolase activity| |circulatory system development| |regulation of nervous system development| |negative regulation of developmental process| |regulation of cellular component biogenesis| |negative regulation of multicellular organismal process| |positive regulation of cellular component organization| |regulation of hydrolase activity| |regulation of organelle organization| |positive regulation of catalytic activity| |positive regulation of molecular function| |regulation of intracellular signal transduction| \\ === CRISPR Data === ^Screen^Score^ |Atovaquone 15μM R08 exp451|-1.82| |Ammonium tetrathiomolybdate 10μM R08 exp448|-1.74| |LY2090314 0.003μM R08 exp500 no dilution day6|1.85| ^Gene^Correlation^ |CCDC64B|0.424| |MEPCE|0.423| |SF3A1|0.423| |POLR2F|0.412| |CHD4|0.412| |RPS2|0.403| |NAPA|0.402| |TCEB3CL2|0.402| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 2047 * **Expression level (log2 read counts)**: 0.02
