This protein bridges B cell receptor-associated kinase activation with downstream signaling pathways, thereby affecting various biological functions. The phosphorylation of five tyrosine residues is necessary for this protein to nucleate distinct signaling effectors following B cell receptor activation. Mutations in this gene cause hypoglobulinemia and absent B cells, a disease in which the pro- to pre-B-cell transition is developmentally blocked. Deficiency in this protein has also been shown in some cases of pre-B acute lymphoblastic leukemia. Alternatively spliced transcript variants have been found for this gene. [provided by RefSeq, May 2012]. COMPLETENESS: complete on the 3' end. * **UniProt Summary**: Functions as a central linker protein, downstream of the B-cell receptor (BCR), bridging the SYK kinase to a multitude of signaling pathways and regulating biological outcomes of B-cell function and development. Plays a role in the activation of ERK/EPHB2, MAP kinase p38 and JNK. Modulates AP1 activation. Important for the activation of NF-kappa-B and NFAT. Plays an important role in BCR-mediated PLCG1 and PLCG2 activation and Ca(2+) mobilization and is required for trafficking of the BCR to late endosomes. However, does not seem to be required for pre-BCR- mediated activation of MAP kinase and phosphatidyl-inositol 3 (PI3) kinase signaling. May be required for the RAC1-JNK pathway. Plays a critical role in orchestrating the pro-B cell to pre-B cell transition. May play an important role in BCR-induced B-cell apoptosis. {ECO:0000269|PubMed:10583958, ECO:0000269|PubMed:15270728, ECO:0000269|PubMed:16912232, ECO:0000269|PubMed:9697839}. |SH2| |transmembrane receptor protein tyrosine kinase adaptor activity| |SH3/SH2 adaptor activity| |cytoplasmic ribonucleoprotein granule| |B cell differentiation| |B cell activation| |lymphocyte differentiation| |leukocyte differentiation| |humoral immune response| |lymphocyte activation| |inflammatory response| |transmembrane receptor protein tyrosine kinase signaling pathway| |hemopoiesis| |hematopoietic or lymphoid organ development| |immune system development| |enzyme linked receptor protein signaling pathway| |leukocyte activation| |cell activation| |defense response| |positive regulation of signal transduction| |intracellular signal transduction| |positive regulation of cell communication| |positive regulation of signaling| |immune response| \\ === CRISPR Data === ^Screen^Score^ |Lead acetate 2000μM R08 exp497|-2.57| |5-Azacytidine 2μM R07 exp330|-2.07| |UMK57 1μM R01 exp59|-2.06| |Quercetin 20μM R08 exp517|-1.95| |Artemisinin 50μM R08 exp450|-1.92| |LY2090314 0.003μM R08 exp499|-1.87| |Dorsomorphin 5μM R07 exp357|-1.84| |Olaparib 4μM R08 exp512|-1.84| |Q15 2.7μM R02 exp79|-1.83| |Lead acetate 2000μM R08 exp498 no dilution day6|-1.8| |Vemurafenib 6.6μM R02 exp89|-1.79| |Diepoxybutane 3μM R07 exp354|-1.74| |Vinorelbine 0.001μM R05 exp249|1.84| |Rapamycin 1μM R00 exp29|1.91| |Rapamycin 10μM R00 exp30|1.93| |INK128 0.2μM R02 exp70|2.04| ^Gene^Correlation^ |BTK|0.6| |RRM1|0.479| |NCK1|0.472| |PLCG2|0.45| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 6272 * **Expression level (log2 read counts)**: 6.8
