The zeta chain plays an important role in coupling antigen recognition to several intracellular signal-transduction pathways. Low expression of the antigen results in impaired immune response. Two alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Part of the TCR-CD3 complex present on T-lymphocyte cell surface that plays an essential role in adaptive immune response. When antigen presenting cells (APCs) activate T-cell receptor (TCR), TCR-mediated signals are transmitted across the cell membrane by the CD3 chains CD3D, CD3E, CD3G and CD3Z. All CD3 chains contain immunoreceptor tyrosine-based activation motifs (ITAMs) in their cytoplasmic domain. Upon TCR engagement, these motifs become phosphorylated by Src family protein tyrosine kinases LCK and FYN, resulting in the activation of downstream signaling pathways (PubMed:2470098, PubMed:7509083). CD3Z ITAMs phosphorylation creates multiple docking sites for the protein kinase ZAP70 leading to ZAP70 phosphorylation and its conversion into a catalytically active enzyme (PubMed:7509083). Plays an important role in intrathymic T-cell differentiation. Additionally, participates in the activity-dependent synapse formation of retinal ganglion cells (RGCs) in both the retina and dorsal lateral geniculate nucleus (dLGN) (By similarity). {ECO:0000250|UniProtKB:P24161, ECO:0000269|PubMed:16027224, ECO:0000269|PubMed:2470098, ECO:0000269|PubMed:28465009, ECO:0000269|PubMed:7509083}. |TCR zetazeta| |ITAM| |alpha-beta T cell receptor complex| |interleukin-2 production| |positive regulation of protein localization to cell surface| |protein tyrosine kinase binding| |regulation of defense response to virus by virus| |protein homotrimerization| |regulation of protein localization to cell surface| |protein trimerization| |protein homotetramerization| |T cell receptor complex| |Fc-gamma receptor signaling pathway involved in phagocytosis| |immune response-regulating cell surface receptor signaling pathway involved in phagocytosis| |transmembrane signaling receptor activity| |Fc receptor mediated stimulatory signaling pathway| |Fc-gamma receptor signaling pathway| |protein tetramerization| |cytokine production| |T cell receptor signaling pathway| |Fc receptor signaling pathway| |antigen receptor-mediated signaling pathway| |positive regulation of cellular protein localization| |phagocytosis| |protein homooligomerization| |immune response-activating cell surface receptor signaling pathway| |immune response-regulating cell surface receptor signaling pathway| |protein complex oligomerization| |regulation of cellular protein localization| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |adaptive immune response| |activation of immune response| |viral process| |symbiotic process| |interspecies interaction between organisms| |positive regulation of immune response| |protein homodimerization activity| |regulation of cellular localization| |regulation of protein localization| |identical protein binding| |immune effector process| |regulation of immune response| |positive regulation of immune system process| |protein-containing complex assembly| |regulation of immune system process| |protein-containing complex subunit organization| |immune response| |vesicle-mediated transport| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp190|Vincristine 0.0005μM R04 exp190]]|-1.71| |[[:results:exp10|CCCP 0.1μM R00 exp10]]|1.76| No correlation found to any other genes in chemogenomics.
