The ceramide synthase enzymes regulate sphingolipid synthesis by catalyzing the formation of ceramides from sphingoid base and acyl-coA substrates. This family member is involved in the synthesis of ceramides with ultra-long-chain acyl moieties (ULC-Cers), important to the epidermis in its role in creating a protective barrier from the environment. The protein encoded by this gene has also been implicated in modification of the lipid structures required for spermatogenesis. Mutations in this gene have been associated with male fertility defects, and epidermal defects, including ichthyosis. Alternative splicing results in multiple transcript variants encoding different isoforms. [provided by RefSeq, Aug 2015]. * **UniProt Summary**: Has (dihydro)ceramide synthesis activity with relatively broad substrate specificity, but a preference for C18:0 and other middle- to long-chain fatty acyl-CoAs (By similarity). It is crucial for the synthesis of very long-chain ceramides in the epidermis, to maintain epidermal lipid homeostasis and terminal differentiation. {ECO:0000250, ECO:0000269|PubMed:23754960}. |TRAM LAG1 CLN8| |Homeobox| |sphingosine N-acyltransferase activity| |ceramide biosynthetic process| |ceramide metabolic process| |sphingolipid biosynthetic process| |membrane lipid biosynthetic process| |sphingolipid metabolic process| |membrane lipid metabolic process| |nuclear membrane| |keratinocyte differentiation| |epidermal cell differentiation| |skin development| |epidermis development| |amide biosynthetic process| |lipid biosynthetic process| |epithelial cell differentiation| |cellular amide metabolic process| |endoplasmic reticulum membrane| |cellular lipid metabolic process| |endoplasmic reticulum| |epithelium development| |lipid metabolic process| |organonitrogen compound biosynthetic process| |DNA binding| |DNA-binding transcription factor activity, RNA polymerase II-specific| |cellular nitrogen compound biosynthetic process| |tissue development| \\ === CRISPR Data === ^Screen^Score^ |Phenformin 20μM R05 exp212|1.7| |Vinblastine 0.2μM R03 exp108|1.76| |Vemurafenib 6.6μM R02 exp89|1.81| |Etoposide 0.1μM R05 exp198|1.97| No correlation found to any other genes in chemogenomics.
