In eukaryotes, CoA and its derivatives are also involved in membrane trafficking and signal transduction. This gene encodes the bifunctional protein coenzyme A synthase (CoAsy) which carries out the last two steps in the biosynthesis of CoA from pantothenic acid (vitamin B5). The phosphopantetheine adenylyltransferase domain of this bifunctional protein catalyzes the conversion of 4'-phosphopantetheine into dephospho-coenzyme A (dpCoA) while its dephospho-CoA kinase domain completes the final step by phosphorylating dpCoA to form CoA. Mutations in this gene are associated with neurodegeneration with brain iron accumulation (NBIA). Alternative splicing results in multiple isoforms. [provided by RefSeq, Apr 2014]. * **UniProt Summary**: Bifunctional enzyme that catalyzes the fourth and fifth sequential steps of CoA biosynthetic pathway. The fourth reaction is catalyzed by the phosphopantetheine adenylyltransferase, coded by the coaD domain; the fifth reaction is catalyzed by the dephospho-CoA kinase, coded by the coaE domain. May act as a point of CoA biosynthesis regulation. {ECO:0000269|PubMed:11923312}. |CoaE| |CTP transf 2| |pantetheine-phosphate adenylyltransferase activity| |dephospho-CoA kinase activity| |coenzyme A biosynthetic process| |coenzyme A metabolic process| |ribonucleoside bisphosphate biosynthetic process| |nucleoside bisphosphate biosynthetic process| |purine nucleoside bisphosphate biosynthetic process| |nucleoside bisphosphate metabolic process| |purine nucleoside bisphosphate metabolic process| |ribonucleoside bisphosphate metabolic process| |coenzyme biosynthetic process| |purine ribonucleotide biosynthetic process| |purine nucleotide biosynthetic process| |ribonucleotide biosynthetic process| |mitochondrial outer membrane| |ribose phosphate biosynthetic process| |purine-containing compound biosynthetic process| |cofactor biosynthetic process| |nucleotide biosynthetic process| |nucleoside phosphate biosynthetic process| |coenzyme metabolic process| |purine ribonucleotide metabolic process| |ribonucleotide metabolic process| |purine nucleotide metabolic process| |ribose phosphate metabolic process| |purine-containing compound metabolic process| |mitochondrial matrix| |cofactor metabolic process| |nucleotide metabolic process| |nucleoside phosphate metabolic process| |organophosphate biosynthetic process| |nucleobase-containing small molecule metabolic process| |carbohydrate derivative biosynthetic process| |organophosphate metabolic process| |carbohydrate derivative metabolic process| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |phosphorylation| |organic cyclic compound biosynthetic process| |organonitrogen compound biosynthetic process| |ATP binding| |cellular nitrogen compound biosynthetic process| |small molecule metabolic process| \\ === CRISPR Data === No hits were found.
