Has endonuclease activity, and functions as mRNA 3'-end-processing endonuclease. Also involved in the histone 3'-end pre-mRNA processing. U7 snRNP- dependent protein that induces both the 3'-endoribonucleolytic cleavage of histone pre-mRNAs and acts as a 5' to 3' exonuclease for degrading the subsequent downstream cleavage product (DCP) of mature histone mRNAs. Cleavage occurs after the 5'-ACCCA-3' sequence in the histone pre-mRNA leaving a 3'hydroxyl group on the upstream fragment containing the stem loop (SL) and 5' phosphate on the downstream cleavage product (DCP) starting with CU nucleotides. The U7-dependent 5' to 3' exonuclease activity is processive and degrades the DCP RNA substrate even after complete removal of the U7-binding site. Binds to the downstream cleavage product (DCP) of histone pre-mRNAs and the cleaved DCP RNA substrate in a U7 snRNP dependent manner. {ECO:0000269|PubMed:14749727, ECO:0000269|PubMed:15037765, ECO:0000269|PubMed:17128255, ECO:0000269|PubMed:18688255}. |Beta-Casp| |RMMBL| |Lactamase B 2| |CPSF73-100 C| |Lactamase B| |mRNA 3-end processing by stem-loop binding and cleavage| |5-3 exonuclease activity| |mRNA cleavage and polyadenylation specificity factor complex| |mRNA cleavage| |endoribonuclease activity| |mRNA polyadenylation| |histone mRNA metabolic process| |RNA polyadenylation| |termination of RNA polymerase II transcription| |DNA-templated transcription, termination| |RNA phosphodiester bond hydrolysis, endonucleolytic| |mRNA 3-end processing| |mRNA export from nucleus| |mRNA-containing ribonucleoprotein complex export from nucleus| |ribonucleoprotein complex export from nucleus| |ribonucleoprotein complex localization| |RNA export from nucleus| |RNA 3-end processing| |protein export from nucleus| |mRNA transport| |nuclear export| |RNA phosphodiester bond hydrolysis| |nucleic acid transport| |RNA transport| |establishment of RNA localization| |RNA localization| |nucleobase-containing compound transport| |protein-containing complex localization| |nucleocytoplasmic transport| |nuclear transport| |mRNA splicing, via spliceosome| |RNA splicing, via transesterification reactions with bulged adenosine as nucleophile| |nucleic acid phosphodiester bond hydrolysis| |RNA splicing, via transesterification reactions| |RNA splicing| |mRNA processing| |transcription by RNA polymerase II| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |mRNA metabolic process| |RNA processing| |intracellular protein transport| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |organic cyclic compound biosynthetic process| |RNA binding| |protein transport| |intracellular transport| |peptide transport| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |cellular nitrogen compound biosynthetic process| |RNA metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |establishment of localization in cell| |nitrogen compound transport| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |2-Deoxy-D-glucose 800μM R05 exp214|-1.95| |Menadione 5μM R06 exp292|1.72| |YM155 0.0002μM R01 exp61|1.77| ^Gene^Correlation^ |HGC6.3|0.437| Global Fraction of Cell Lines Where Essential: 492/726 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|1/1| |909776.0|1/1| |bile duct|20/28| |blood|18/28| |bone|20/25| |breast|20/33| |central nervous system|43/56| |cervix|4/4| |colorectal|11/17| |esophagus|11/13| |fibroblast|1/1| |gastric|10/15| |kidney|14/21| |liver|11/20| |lung|52/75| |lymphocyte|13/14| |ovary|16/26| |pancreas|18/24| |peripheral nervous system|14/16| |plasma cell|12/15| |prostate|1/1| |skin|13/24| |soft tissue|5/7| |thyroid|2/2| |upper aerodigestive|15/22| |urinary tract|17/29| |uterus|4/5| == Essentiality in NALM6 == * **Essentiality Rank**: 211 * **Expression level (log2 read counts)**: 6.46
