The protein encoded by this gene is the fourth, or delta, subunit. Defects in this gene are a cause of leukoencephalopathy with vanishing white matter (VWM) and ovarioleukodystrophy. Multiple transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Catalyzes the exchange of eukaryotic initiation factor 2-bound GDP for GTP. |IF-2B| |eukaryotic translation initiation factor 2B complex| |translation initiation factor binding| |oligodendrocyte development| |ovarian follicle development| |translation initiation factor activity| |oligodendrocyte differentiation| |female gonad development| |development of primary female sexual characteristics| |guanyl-nucleotide exchange factor activity| |myelination| |glial cell development| |female sex differentiation| |ensheathment of neurons| |axon ensheathment| |response to heat| |response to glucose| |translational initiation| |response to hexose| |response to monosaccharide| |glial cell differentiation| |response to carbohydrate| |T cell receptor signaling pathway| |response to temperature stimulus| |gonad development| |gliogenesis| |development of primary sexual characteristics| |sex differentiation| |antigen receptor-mediated signaling pathway| |regulation of translation| |translation| |response to peptide hormone| |regulation of cellular amide metabolic process| |peptide biosynthetic process| |reproductive structure development| |reproductive system development| |immune response-activating cell surface receptor signaling pathway| |response to peptide| |immune response-regulating cell surface receptor signaling pathway| |posttranscriptional regulation of gene expression| |amide biosynthetic process| |peptide metabolic process| |immune response-activating signal transduction| |immune response-regulating signaling pathway| |activation of immune response| |developmental process involved in reproduction| |cellular amide metabolic process| |positive regulation of immune response| |response to hormone| |central nervous system development| |response to organonitrogen compound| |response to nitrogen compound| |positive regulation of immune system process| |regulation of immune response| |response to abiotic stimulus| |organonitrogen compound biosynthetic process| |reproductive process| |reproduction| |response to endogenous stimulus| |response to oxygen-containing compound| |neurogenesis| |cellular nitrogen compound biosynthetic process| |cell development| |regulation of immune system process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |AICAR 240μM R03 exp116|-1.78| |Metformin 100 to 150μM on day4 R04 exp170|1.7| |Suberoylanilide-Hydroxamic-Acid 0.02μM R01 exp53|1.71| |GSK-J4 1μM R07 exp362|1.72| |Taxol 0.002μM R01 exp54|1.74| |IU1-C 25μM R04 exp183|1.76| |Losmapimod 1μM R07 exp376|1.78| |BI-6727 0.0125μM R03 exp97|1.83| |Deguelin 0.15μM R05 exp209|1.83| |35°C R06 exp302|1.86| |MLN-4924 0.2μM R00 exp21|1.91| |2-Methoxyestradiol 0.55μM R07 exp310|2.12| |S-trityl-L-cysteine 0.5μM R05 exp243|2.18| |Chloramphenicol 2μM R00 exp12|2.3| ^Gene^Correlation^ |PPP1R15B|0.477| |EIF2B5|0.415| |DNAJC16|0.412| Global Fraction of Cell Lines Where Essential: 297/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|8/28| |blood|11/28| |bone|9/26| |breast|12/33| |central nervous system|24/56| |cervix|1/4| |colorectal|9/17| |esophagus|6/13| |fibroblast|0/1| |gastric|10/16| |kidney|12/21| |liver|6/20| |lung|29/75| |lymphocyte|11/16| |ovary|10/26| |pancreas|6/24| |peripheral nervous system|9/16| |plasma cell|6/15| |prostate|1/1| |skin|4/24| |soft tissue|5/9| |thyroid|1/2| |upper aerodigestive|7/22| |urinary tract|12/29| |uterus|2/5| == Essentiality in NALM6 == * **Essentiality Rank**: 700 * **Expression level (log2 read counts)**: 5.98
