The eIF-3 complex associates with the 40S ribosome and facilitates the recruitment of eIF-1, eIF-1A, eIF-2:GTP:methionyl-tRNAi and eIF-5 to form the 43S pre- initiation complex (43S PIC). The eIF-3 complex stimulates mRNA recruitment to the 43S PIC and scanning of the mRNA for AUG recognition. The eIF-3 complex is also required for disassembly and recycling of post-termination ribosomal complexes and subsequently prevents premature joining of the 40S and 60S ribosomal subunits prior to initiation (PubMed:17581632). The eIF- 3 complex specifically targets and initiates translation of a subset of mRNAs involved in cell proliferation, including cell cycling, differentiation and apoptosis, and uses different modes of RNA stem-loop binding to exert either translational activation or repression (PubMed:25849773). {ECO:0000255|HAMAP-Rule:MF_03008, ECO:0000269|PubMed:17581632, ECO:0000269|PubMed:25849773, ECO:0000269|PubMed:27462815}. |WD40| |eukaryotic translation initiation factor 3 complex, eIF3m| |eukaryotic 48S preinitiation complex| |formation of cytoplasmic translation initiation complex| |eukaryotic translation initiation factor 3 complex| |eukaryotic 43S preinitiation complex| |cytoplasmic translational initiation| |translation initiation factor activity| |cytoplasmic translation| |translational initiation| |ribonucleoprotein complex assembly| |ribonucleoprotein complex subunit organization| |translation| |peptide biosynthetic process| |ribonucleoprotein complex biogenesis| |amide biosynthetic process| |peptide metabolic process| |cellular amide metabolic process| |cellular protein-containing complex assembly| |organonitrogen compound biosynthetic process| |protein-containing complex assembly| |cellular nitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |protein-containing complex subunit organization| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Rotenone 20μM R00 exp34|1.82| |Etoposide 1μM R00 exp19|1.83| |MLN-4924 2μM R00 exp22|1.84| |IU1-C 25μM R04 exp183|1.94| |Bortezomib 0.05μM R00 exp7|1.97| |SB743921 0.001μM R05 exp244|2.03| |Oligomycin-A 20μM R00 exp26|2.06| |TRAIL 50ng/ml R00 exp36|2.17| |Etoposide 0.3μM R05 exp199|2.35| No correlation found to any other genes in chemogenomics.
