Binding to FUSE occurs on the non-coding strand, and is important to the regulation of c-myc in undifferentiated cells. This protein contains three domains, an amphipathic helix N-terminal domain, a DNA-binding central domain, and a C-terminal transactivation domain that contains three tyrosine-rich motifs. The N-terminal domain is thought to repress the activity of the C-terminal domain. This protein is also thought to bind RNA, and contains 3'-5' helicase activity with in vitro activity on both DNA-DNA and RNA-RNA duplexes. Aberrant expression of this gene has been found in malignant tissues, and this gene is important to neural system and lung development. Binding of this protein to viral RNA is thought to play a role in several viral diseases, including hepatitis C and hand, foot and mouth disease. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Dec 2014]. * **UniProt Summary**: Regulates MYC expression by binding to a single-stranded far-upstream element (FUSE) upstream of the MYC promoter. May act both as activator and repressor of transcription. {ECO:0000269|PubMed:8125259}. |KH 2| |DUF1897| |KH 1| |single-stranded DNA binding| |transcription by RNA polymerase II| |transcription, DNA-templated| |nucleic acid-templated transcription| |RNA biosynthetic process| |DNA-binding transcription factor activity| |nucleobase-containing compound biosynthetic process| |heterocycle biosynthetic process| |aromatic compound biosynthetic process| |organic cyclic compound biosynthetic process| |RNA binding| |cellular nitrogen compound biosynthetic process| |RNA metabolic process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |positive regulation of gene expression| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |2-Methoxyestradiol 0.55 to 1μM on day4 R07 exp312|1.71| |Cerivastatin 0.15μM R05 exp226|1.9| |Fluvastatin 2.2μM R06 exp282|2.09| ^Gene^Correlation^ |RRM1|0.448| Global Fraction of Cell Lines Where Essential: 1/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|1/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 498 * **Expression level (log2 read counts)**: 8.23
