This gene encodes a receptor-independent activator of G protein signaling, which is one of several factors that influence the basal activity of G-protein signaling systems. The protein contains seven tetratricopeptide repeats in its N-terminal half and four G-protein regulatory (GPR) motifs in its C-terminal half. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Dec 2011]. * **UniProt Summary**: Guanine nucleotide dissociation inhibitor (GDI) which functions as a receptor-independent activator of heterotrimeric G- protein signaling. Keeps G(i/o) alpha subunit in its GDP-bound form thus uncoupling heterotrimeric G-proteins signaling from G protein-coupled receptors. Controls spindle orientation and asymmetric cell fate of cerebral cortical progenitors. May also be involved in macroautophagy in intestinal cells. May play a role in drug addiction. {ECO:0000269|PubMed:11024022, ECO:0000269|PubMed:12642577}. |GoLoco| |negative regulation of guanyl-nucleotide exchange factor activity| |regulation of guanyl-nucleotide exchange factor activity| |GDP-dissociation inhibitor activity| |regulation of GTP binding| |negative regulation of GTPase activity| |positive regulation of macroautophagy| |negative regulation of protein binding| |positive regulation of autophagy| |cell cortex| |negative regulation of binding| |regulation of macroautophagy| |positive regulation of binding| |regulation of protein binding| |regulation of autophagy| |positive regulation of cellular catabolic process| |regulation of binding| |positive regulation of catabolic process| |negative regulation of hydrolase activity| |regulation of GTPase activity| |protein-containing complex| |Golgi membrane| |negative regulation of catalytic activity| |regulation of cellular catabolic process| |endoplasmic reticulum membrane| |regulation of catabolic process| |negative regulation of molecular function| |regulation of hydrolase activity| |positive regulation of molecular function| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp48|Mubritinib 0.2μM R01 exp48]]|1.77| |[[:results:exp40|2-Methoxyestradiol 0.2μM R01 exp40]]|1.93| No correlation found to any other genes in chemogenomics.
