This enzyme is a homodimeric flavoprotein. It is a central enzyme of cellular antioxidant defense, and reduces oxidized glutathione disulfide (GSSG) to the sulfhydryl form GSH, which is an important cellular antioxidant. Rare mutations in this gene result in hereditary glutathione reductase deficiency. Multiple alternatively spliced transcript variants encoding different isoforms have been found. [provided by RefSeq, Aug 2010]. * **UniProt Summary**: Maintains high levels of reduced glutathione in the cytosol. |Pyr redox| |Pyr redox 2| |FAD oxidored| |Pyr redox dim| |glutathione-disulfide reductase activity| |nucleobase-containing small molecule interconversion| |NADP binding| |glutathione metabolic process| |flavin adenine dinucleotide binding| |electron transfer activity| |cell redox homeostasis| |cellular oxidant detoxification| |cellular detoxification| |detoxification| |electron transport chain| |cellular modified amino acid metabolic process| |cellular response to toxic substance| |cellular response to oxidative stress| |sulfur compound metabolic process| |mitochondrial matrix| |response to oxidative stress| |external side of plasma membrane| |generation of precursor metabolites and energy| |cofactor metabolic process| |response to toxic substance| |peptide metabolic process| |nucleobase-containing small molecule metabolic process| |cellular amide metabolic process| |cellular homeostasis| |oxidation-reduction process| |mitochondrion| |homeostatic process| |cellular response to stress| |small molecule metabolic process| \\ === CRISPR Data === ^Screen^Score^ |Celastrol 0.12μM R05 exp225|-2.83| |QNZ 0.01μM R05 exp241|-2.45| |Arsenate 40μM R06 exp264|-1.96| |A-395 10μM R05 exp218|-1.87| |Methotrexate 0.01μM R08 exp501|1.86| ^Gene^Correlation^ |RRM1|0.427| Global Fraction of Cell Lines Where Essential: 1/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|1/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 12254 * **Expression level (log2 read counts)**: 6.15
