This gene encodes the interleukin 1 receptor accessory protein. The protein is a necessary part of the interleukin 1 receptor complex which initiates signalling events that result in the activation of interleukin 1-responsive genes. Alternative splicing of this gene results in two transcript variants encoding two different isoforms, one membrane-bound and one soluble. The ratio of soluble to membrane-bound forms increases during acute-phase induction or stress. [provided by RefSeq, Nov 2009]. * **UniProt Summary**: Coreceptor for IL1RL2 in the IL-36 signaling system (By similarity). Coreceptor with IL1R1 in the IL-1 signaling system. Associates with IL1R1 bound to IL1B to form the high affinity interleukin-1 receptor complex which mediates interleukin-1- dependent activation of NF-kappa-B and other pathways. Signaling involves the recruitment of adapter molecules such as TOLLIP, MYD88, and IRAK1 or IRAK2 via the respective TIR domains of the receptor/coreceptor subunits. Recruits TOLLIP to the signaling complex. Does not bind to interleukin-1 alone; binding of IL1RN to IL1R1, prevents its association with IL1R1 to form a signaling complex. The cellular response is modulated through a non- signaling association with the membrane IL1R2 decoy receptor. Coreceptor for IL1RL1 in the IL-33 signaling system. Can bidirectionally induce pre- and postsynaptic differentiation of neurons by trans-synaptically binding to PTPRD (By similarity). {ECO:0000250|UniProtKB:Q61730, ECO:0000269|PubMed:10799889, ECO:0000269|PubMed:9371760, ECO:0000305|PubMed:19836339}. Isoform 4: Unable to mediate canonical IL-1 signaling (PubMed:19481478). Required for Src phosphorylation by IL1B. May be involved in IL1B-potentiated NMDA-induced calcium influx in neurons (By similarity). {ECO:0000250|UniProtKB:Q61730, ECO:0000269|PubMed:19481478}. |TIR| |interleukin-33 receptor activity| |interleukin-33-mediated signaling pathway| |interleukin-2 biosynthetic process| |interleukin-4 secretion| |interleukin-1 receptor activity| |interleukin-4 production| |trans-synaptic signaling by trans-synaptic complex| |interleukin-2 production| |regulation of postsynaptic density assembly| |regulation of postsynapse assembly| |regulation of postsynaptic specialization assembly| |positive regulation of interleukin-5 production| |interleukin-1 receptor binding| |regulation of excitatory synapse assembly| |cytokine biosynthetic process| |regulation of postsynaptic density organization| |cytokine metabolic process| |positive regulation of interleukin-13 production| |regulation of interleukin-13 production| |regulation of interleukin-5 production| |synaptic membrane adhesion| |regulation of presynapse assembly| |regulation of presynapse organization| |positive regulation of interleukin-6 secretion| |cytokine secretion| |positive regulation of synapse assembly| |positive regulation of interleukin-6 production| |interleukin-1-mediated signaling pathway| |regulation of postsynapse organization| |regulation of synapse assembly| |positive regulation of cytokine secretion| |regulation of interleukin-6 production| |cytokine production| |positive regulation of NF-kappaB transcription factor activity| |protein secretion| |establishment of protein localization to extracellular region| |protein localization to extracellular region| |peptide secretion| |cellular response to interleukin-1| |response to interleukin-1| |regulation of cytokine secretion| |regulation of synapse organization| |regulation of organelle assembly| |regulation of synapse structure or activity| |cell-cell adhesion via plasma-membrane adhesion molecules| |positive regulation of DNA-binding transcription factor activity| |synapse organization| |positive regulation of protein secretion| |positive regulation of peptide secretion| |glutamatergic synapse| |positive regulation of secretion by cell| |positive regulation of protein transport| |regulation of DNA-binding transcription factor activity| |positive regulation of secretion| |trans-synaptic signaling| |positive regulation of cytokine production| |positive regulation of establishment of protein localization| |synaptic signaling| |regulation of protein secretion| |regulation of peptide secretion| |inflammatory response| |cell-cell adhesion| |positive regulation of cellular component biogenesis| |positive regulation of nervous system development| |cytokine-mediated signaling pathway| |regulation of cytokine production| |regulation of protein transport| |regulation of peptide transport| |regulation of establishment of protein localization| |regulation of secretion by cell| |innate immune response| |regulation of secretion| |regulation of nervous system development| |cell adhesion| |biological adhesion| |defense response to other organism| |regulation of cellular component biogenesis| |positive regulation of transport| |secretion by cell| |cellular response to cytokine stimulus| |regulation of protein localization| |export from cell| |response to cytokine| |cell-cell signaling| |secretion| |positive regulation of cellular component organization| |regulation of organelle organization| |response to other organism| |response to external biotic stimulus| |response to biotic stimulus| |defense response| |positive regulation of developmental process| |integral component of plasma membrane| |organonitrogen compound biosynthetic process| |protein transport| |peptide transport| |protein-containing complex assembly| |amide transport| |establishment of protein localization| |positive regulation of multicellular organismal process| |macromolecule biosynthetic process| |positive regulation of molecular function| |nitrogen compound transport| |protein-containing complex subunit organization| |regulation of transport| |immune response| |extracellular region| |membrane| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp372|Ibrutinib 1μM R07 exp372]]|-2.05| |[[:results:exp262|Alda-1 10μM R06 exp262]]|1.93| No correlation found to any other genes in chemogenomics.
