The encoded protein is localized at the centrosome and interacts with other centrosome components as part of a multiprotein complex that regulates dynein function. This protein plays an essential role in microtubule organization, mitosis and neuronal migration. Mutations in this gene cause lissencephaly 4, a disorder characterized by lissencephaly, severe brain atrophy, microcephaly, and severe cognitive disability. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Mar 2012]. * **UniProt Summary**: Required for centrosome duplication and formation and function of the mitotic spindle. Essential for the development of the cerebral cortex. May regulate the production of neurons by controlling the orientation of the mitotic spindle during division of cortical neuronal progenitors of the proliferative ventricular zone of the brain. Orientation of the division plane perpendicular to the layers of the cortex gives rise to two proliferative neuronal progenitors whereas parallel orientation of the division plane yields one proliferative neuronal progenitor and a post- mitotic neuron. A premature shift towards a neuronal fate within the progenitor population may result in an overall reduction in the final number of neurons and an increase in the number of neurons in the deeper layers of the cortex. {ECO:0000269|PubMed:17600710, ECO:0000269|PubMed:21529752}. |NUDE C| |mitotic centrosome separation| |centrosome separation| |regulation of microtubule motor activity| |spindle pole centrosome| |microtubule nucleation| |establishment of mitotic spindle orientation| |centrosome localization| |centrosome duplication| |microtubule organizing center localization| |neuroblast proliferation| |microtubule polymerization| |establishment of mitotic spindle localization| |establishment of spindle orientation| |establishment of spindle localization| |vesicle transport along microtubule| |spindle localization| |vesicle cytoskeletal trafficking| |microtubule polymerization or depolymerization| |stem cell proliferation| |kinesin complex| |cleavage furrow| |establishment of chromosome localization| |chromosome localization| |neural precursor cell proliferation| |organelle transport along microtubule| |centrosome cycle| |kinetochore| |protein polymerization| |condensed chromosome kinetochore| |microtubule organizing center organization| |ciliary basal body-plasma membrane docking| |microtubule cytoskeleton organization involved in mitosis| |cerebral cortex development| |establishment of cell polarity| |neuron migration| |G2/M transition of mitotic cell cycle| |cell cycle G2/M phase transition| |mitotic nuclear division| |transport along microtubule| |organelle localization by membrane tethering| |pallium development| |establishment of vesicle localization| |cytoskeleton-dependent intracellular transport| |membrane docking| |microtubule-based transport| |vesicle localization| |regulation of G2/M transition of mitotic cell cycle| |establishment or maintenance of cell polarity| |regulation of cell cycle G2/M phase transition| |microtubule binding| |protein domain specific binding| |telencephalon development| |synapse| |mitotic cell cycle phase transition| |microtubule-based movement| |chromosome segregation| |cell cycle phase transition| |nuclear division| |organelle fission| |microtubule| |cilium assembly| |establishment of organelle localization| |cilium organization| |forebrain development| |regulation of mitotic cell cycle phase transition| |plasma membrane bounded cell projection assembly| |regulation of cell cycle phase transition| |supramolecular fiber organization| |cell projection assembly| |microtubule cytoskeleton organization| |centrosome| |cell division| |cell population proliferation| |organelle localization| |mitotic cell cycle process| |regulation of mitotic cell cycle| |microtubule-based process| |mitotic cell cycle| |brain development| |regulation of cell cycle process| |organelle assembly| |head development| |cellular protein-containing complex assembly| |cell migration| |central nervous system development| |cell cycle process| |identical protein binding| |localization of cell| |cell motility| |cytoskeleton organization| |plasma membrane bounded cell projection organization| |cell projection organization| |regulation of cell cycle| |regulation of hydrolase activity| |locomotion| |cell cycle| |intracellular transport| |generation of neurons| |movement of cell or subcellular component| |protein-containing complex assembly| |neurogenesis| |establishment of localization in cell| |protein-containing complex subunit organization| |membrane| \\ === CRISPR Data === ^Screen^Score^ |2-Methoxyestradiol 0.55μM R07 exp310|1.77| |Combretastatin A4 0.002 to 0.003μM day4 R04 exp179|1.77| |SB743921 0.001μM R05 exp244|2.14| |Vinblastine 0.002μM R01 exp60|2.41| |2-Methoxyestradiol 0.55 to 1μM on day4 R07 exp312|2.43| |UMK57 1μM R01 exp59|2.6| |Colchicine 0.009μM R05 exp215|2.62| |BML-284 0.09μM R08 exp460|3.14| |UM131593 0.2μM R03 exp106|3.23| |S-trityl-L-cysteine 0.5μM R05 exp243|3.27| ^Gene^Correlation^ |ATAT1|0.425| Global Fraction of Cell Lines Where Essential: 59/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|1/1| |bile duct|2/28| |blood|2/28| |bone|0/26| |breast|2/33| |central nervous system|2/56| |cervix|1/4| |colorectal|4/17| |esophagus|2/13| |fibroblast|0/1| |gastric|0/16| |kidney|3/21| |liver|2/20| |lung|2/75| |lymphocyte|1/16| |ovary|2/26| |pancreas|7/24| |peripheral nervous system|3/16| |plasma cell|0/15| |prostate|0/1| |skin|1/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|1/22| |urinary tract|5/29| |uterus|1/5| == Essentiality in NALM6 == * **Essentiality Rank**: 4244 * **Expression level (log2 read counts)**: 5.4
