The protein forms disulfide-linked oligomers in the extracellular matrix. Mutations in this gene result in Norrie disease and X-linked exudative vitreoretinopathy. [provided by RefSeq, Feb 2009]. * **UniProt Summary**: Activates the canonical Wnt signaling pathway through FZD4 and LRP5 coreceptor. Plays a central role in retinal vascularization by acting as a ligand for FZD4 that signals via stabilizing beta-catenin (CTNNB1) and activating LEF/TCF-mediated transcriptional programs. Acts in concert with TSPAN12 to activate FZD4 independently of the Wnt-dependent activation of FZD4, suggesting the existence of a Wnt-independent signaling that also promote accumulation the beta-catenin (CTNNB1). May be involved in a pathway that regulates neural cell differentiation and proliferation. Possible role in neuroectodermal cell-cell interaction. |Cys knot| |Norrin signaling pathway| |extracellular matrix-cell signaling| |retina vasculature morphogenesis in camera-type eye| |retina vasculature development in camera-type eye| |frizzled binding| |canonical Wnt signaling pathway| |vacuole organization| |retina development in camera-type eye| |placenta development| |cytokine activity| |visual perception| |sensory perception of light stimulus| |positive regulation of DNA-binding transcription factor activity| |camera-type eye development| |cell-cell signaling by wnt| |Wnt signaling pathway| |eye development| |visual system development| |collagen-containing extracellular matrix| |sensory system development| |reproductive structure development| |cell surface receptor signaling pathway involved in cell-cell signaling| |regulation of DNA-binding transcription factor activity| |reproductive system development| |vasculature development| |cardiovascular system development| |sensory organ development| |cell surface| |developmental process involved in reproduction| |circulatory system development| |protein homodimerization activity| |sensory perception| |cell-cell signaling| |nervous system process| |reproductive process| |reproduction| |positive regulation of transcription, DNA-templated| |extracellular space| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |positive regulation of RNA metabolic process| |positive regulation of molecular function| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |system process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === No hits were found.
