Studies in yeast demonstrated that ORC binds specifically to origins of replication and serves as a platform for the assembly of additional initiation factors such as Cdc6 and Mcm proteins. The protein encoded by this gene is a subunit of the ORC complex. Alternatively spliced transcript variants encoding distinct isoforms have been described. [provided by RefSeq, Oct 2010]. * **UniProt Summary**: Component of the origin recognition complex (ORC) that binds origins of replication. DNA-binding is ATP-dependent. The specific DNA sequences that define origins of replication have not been identified yet. ORC is required to assemble the pre- replication complex necessary to initiate DNA replication. No Pfam Domain information is available for this gene. |origin recognition complex| |nuclear origin of replication recognition complex| |DNA replication origin binding| |DNA replication initiation| |nucleotide binding| |nuclear chromosome, telomeric region| |G1/S transition of mitotic cell cycle| |DNA-dependent DNA replication| |cell cycle G1/S phase transition| |DNA replication| |mitotic cell cycle phase transition| |cell cycle phase transition| |mitotic cell cycle process| |mitotic cell cycle| |DNA metabolic process| |molecular function| |cell cycle process| |cell cycle| |ATP binding| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |Ribavirin 10μM R01 exp52|-2.49| |Bisphenol S 100μM R08 exp458|-1.81| |THZ1 0.06μM R08 exp531|1.98| ^Gene^Correlation^ |TBCA|0.42| |PMM2|0.414| |HARS|0.41| |UHRF1|0.41| |ALG2|0.407| |TRMT5|0.407| Global Fraction of Cell Lines Where Essential: 11/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|2/28| |bone|1/26| |breast|1/33| |central nervous system|2/56| |cervix|0/4| |colorectal|1/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|1/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 4946 * **Expression level (log2 read counts)**: 5.05
