The enzyme (cofactors iron and ascorbate) catalyzes the hydroxylation of lysyl residues in collagen-like peptides. The resultant hydroxylysyl groups are attachment sites for carbohydrates in collagen and thus are critical for the stability of intermolecular crosslinks. Some patients with Ehlers-Danlos syndrome type VI have deficiencies in lysyl hydroxylase activity. Two transcript variants encoding different isoforms have been found for this gene. [provided by RefSeq, Oct 2015]. * **UniProt Summary**: Part of a complex composed of PLOD1, P3H3 and P3H4 that catalyzes hydroxylation of lysine residues in collagen alpha chains and is required for normal assembly and cross-linkling of collagen fibrils (By similarity). Forms hydroxylysine residues in -Xaa-Lys-Gly- sequences in collagens (PubMed:8621606, PubMed:10686424, PubMed:15854030). These hydroxylysines serve as sites of attachment for carbohydrate units and are essential for the stability of the intermolecular collagen cross-links (Probable). {ECO:0000250|UniProtKB:Q9R0E2, ECO:0000269|PubMed:10686424, ECO:0000269|PubMed:15854030, ECO:0000269|PubMed:8621606, ECO:0000305}. |2OG-FeII Oxy| |procollagen-lysine 5-dioxygenase activity| |hydroxylysine metabolic process| |hydroxylysine biosynthetic process| |procollagen glucosyltransferase activity| |catalytic complex| |peptidyl-lysine hydroxylation| |rough endoplasmic reticulum membrane| |L-ascorbic acid binding| |aspartate family amino acid biosynthetic process| |protein hydroxylation| |cellular modified amino acid biosynthetic process| |collagen fibril organization| |aspartate family amino acid metabolic process| |collagen metabolic process| |alpha-amino acid biosynthetic process| |cellular amino acid biosynthetic process| |protein O-linked glycosylation| |iron ion binding| |cellular modified amino acid metabolic process| |alpha-amino acid metabolic process| |protein glycosylation| |macromolecule glycosylation| |glycosylation| |carboxylic acid biosynthetic process| |organic acid biosynthetic process| |cellular amino acid metabolic process| |peptidyl-lysine modification| |glycoprotein biosynthetic process| |extracellular matrix organization| |response to hypoxia| |response to decreased oxygen levels| |response to oxygen levels| |glycoprotein metabolic process| |extracellular structure organization| |epidermis development| |supramolecular fiber organization| |small molecule biosynthetic process| |carbohydrate derivative biosynthetic process| |protein homodimerization activity| |peptidyl-amino acid modification| |carboxylic acid metabolic process| |endoplasmic reticulum membrane| |oxidation-reduction process| |oxoacid metabolic process| |organic acid metabolic process| |carbohydrate derivative metabolic process| |response to abiotic stimulus| |organonitrogen compound biosynthetic process| |cellular macromolecule biosynthetic process| |small molecule metabolic process| |tissue development| |macromolecule biosynthetic process| |protein-containing complex subunit organization| \\ === CRISPR Data === ^Screen^Score^ |SB202190 10μM R03 exp148|-1.78| |Senexin-A 25μM R07 exp400|-1.72| |FTY720 3μM R08 exp483|1.73| No correlation found to any other genes in chemogenomics.
