PP2C family members are known to be negative regulators of cell stress response pathways. This phosphatase has been shown to dephosphorylate cyclin-dependent kinases (CDKs), and thus may be involved in cell cycle control. Overexpression of this phosphatase is reported to cause cell-growth arrest or cell death. Alternative splicing results in multiple transcript variants encoding different isoforms. Additional transcript variants have been described, but currently do not represent full-length sequences. [provided by RefSeq, Jul 2008]. * **UniProt Summary**: Enzyme with a broad specificity. Dephosphorylates CDK2 and CDK6 in vitro. Dephosphorylates PRKAA1 and PRKAA2. Inhibits TBK1-mediated antiviral signaling by dephosphorylating it at 'Ser- 172'. Plays an important role in the termination of TNF-alpha- mediated NF-kappa-B activation through dephosphorylating and inactivating IKBKB/IKKB. {ECO:0000269|PubMed:18930133, ECO:0000269|PubMed:22750291}. |PP2C C| |PP2C| |protein myristoylation| |N-terminal protein myristoylation| |N-terminal protein lipidation| |negative regulation of interferon-beta production| |peptidyl-threonine dephosphorylation| |magnesium-dependent protein serine/threonine phosphatase activity| |negative regulation of NIK/NF-kappaB signaling| |negative regulation of defense response to virus| |N-terminal protein amino acid modification| |negative regulation of I-kappaB kinase/NF-kappaB signaling| |negative regulation of type I interferon production| |regulation of interferon-beta production| |manganese ion binding| |protein serine/threonine phosphatase activity| |regulation of defense response to virus| |protein lipidation| |lipoprotein biosynthetic process| |negative regulation of response to biotic stimulus| |regulation of NIK/NF-kappaB signaling| |lipoprotein metabolic process| |negative regulation of immune effector process| |regulation of type I interferon production| |protein acylation| |negative regulation of defense response| |protein dephosphorylation| |magnesium ion binding| |negative regulation of multi-organism process| |regulation of I-kappaB kinase/NF-kappaB signaling| |negative regulation of cytokine production| |dephosphorylation| |negative regulation of response to external stimulus| |negative regulation of immune system process| |regulation of immune effector process| |negative regulation of intracellular signal transduction| |regulation of response to biotic stimulus| |regulation of cytokine production| |regulation of defense response| |regulation of multi-organism process| |nucleolus| |regulation of response to external stimulus| |negative regulation of multicellular organismal process| |negative regulation of signal transduction| |negative regulation of cell communication| |negative regulation of signaling| |organonitrogen compound biosynthetic process| |regulation of response to stress| |negative regulation of response to stimulus| |regulation of immune system process| |cellular macromolecule biosynthetic process| |macromolecule biosynthetic process| |regulation of intracellular signal transduction| |membrane| \\ === CRISPR Data === ^Screen^Score^ |LB-100 4.1μM R02 exp72|-1.95| |2-Methoxyestradiol 0.2μM R01 exp40|1.7| |Rapamycin 0.001μM R08 exp429|1.7| |Bafilomycin-A1 0.009μM R08 exp454|1.76| |EPZ-5676 30μM R05 exp233|2.06| |Doxycycline 10μM R00 exp18|2.22| No correlation found to any other genes in chemogenomics.
