Mutations in this gene have been associated with dystonia. Alternative splicing results in multiple transcript variants. [provided by RefSeq, Nov 2008]. * **UniProt Summary**: Activates EIF2AK2/PKR in the absence of double-stranded RNA (dsRNA), leading to phosphorylation of EIF2S1/EFI2-alpha and inhibition of translation and induction of apoptosis. Required for siRNA production by DICER1 and for subsequent siRNA-mediated post- transcriptional gene silencing. Does not seem to be required for processing of pre-miRNA to miRNA by DICER1. Promotes UBC9-p53/TP53 association and sumoylation and phosphorylation of p53/TP53 at 'Lys-386' at 'Ser-392' respectively and enhances its activity in a EIF2AK2/PKR-dependent manner (By similarity). {ECO:0000250, ECO:0000269|PubMed:10336432, ECO:0000269|PubMed:11238927, ECO:0000269|PubMed:16424907, ECO:0000269|PubMed:16982605, ECO:0000269|PubMed:17452327, ECO:0000269|PubMed:9687506}. |dsrm| |pre-miRNA binding| |RISC-loading complex| |outer ear morphogenesis| |production of siRNA involved in RNA interference| |pre-miRNA processing| |RNA interference| |middle ear morphogenesis| |miRNA metabolic process| |production of miRNAs involved in gene silencing by miRNA| |production of small RNA involved in gene silencing by RNA| |dsRNA processing| |enzyme activator activity| |gene silencing by miRNA| |posttranscriptional gene silencing by RNA| |posttranscriptional gene silencing| |positive regulation of intrinsic apoptotic signaling pathway| |double-stranded RNA binding| |gene silencing by RNA| |ear morphogenesis| |gene silencing| |regulation of intrinsic apoptotic signaling pathway| |positive regulation of apoptotic signaling pathway| |protein stabilization| |ear development| |regulation of gene expression, epigenetic| |skeletal system morphogenesis| |cellular response to oxidative stress| |sensory organ morphogenesis| |response to virus| |regulation of protein stability| |embryonic organ morphogenesis| |enzyme binding| |ncRNA processing| |response to oxidative stress| |regulation of apoptotic signaling pathway| |embryonic organ development| |ncRNA metabolic process| |skeletal system development| |posttranscriptional regulation of gene expression| |sensory organ development| |embryonic morphogenesis| |positive regulation of apoptotic process| |positive regulation of programmed cell death| |negative regulation of cell population proliferation| |perinuclear region of cytoplasm| |positive regulation of cell death| |viral process| |symbiotic process| |interspecies interaction between organisms| |RNA processing| |protein homodimerization activity| |animal organ morphogenesis| |protein phosphorylation| |embryo development| |positive regulation of intracellular signal transduction| |identical protein binding| |phosphorylation| |response to other organism| |response to external biotic stimulus| |response to biotic stimulus| |RNA binding| |positive regulation of catalytic activity| |regulation of apoptotic process| |regulation of programmed cell death| |regulation of cell population proliferation| |positive regulation of signal transduction| |RNA metabolic process| |regulation of cell death| |cellular response to stress| |negative regulation of gene expression| |positive regulation of molecular function| |positive regulation of cell communication| |positive regulation of signaling| |regulation of intracellular signal transduction| |immune response| |membrane| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |[[:results:exp376|Losmapimod 1μM R07 exp376]]|1.73| |[[:results:exp77|Prochlorperazine 5.2μM R02 exp77]]|1.75| No correlation found to any other genes in chemogenomics.
