This protein can activate small GTPases, including RAS and RAP1/RAS3. The nucleotide exchange activity of this protein can be stimulated by calcium and diacylglycerol. Four alternatively spliced transcript variants encoding two different isoforms have been found for this gene. [provided by RefSeq, Jan 2016]. * **UniProt Summary**: Functions as a calcium- and DAG-regulated nucleotide exchange factor specifically activating Rap through the exchange of bound GDP for GTP. May also activates other GTPases such as RRAS, RRAS2, NRAS, KRAS but not HRAS. Functions in aggregation of platelets and adhesion of T-lymphocytes and neutrophils probably through inside-out integrin activation. May function in the muscarinic acetylcholine receptor M1/CHRM1 signaling pathway. {ECO:0000269|PubMed:10918068, ECO:0000269|PubMed:14702343, ECO:0000269|PubMed:17576779, ECO:0000269|PubMed:17702895, ECO:0000269|PubMed:24958846, ECO:0000269|PubMed:27235135}. |RasGEF N| |C1 1| |RasGEF| |efhand| |diacylglycerol binding| |cellular response to calcium ion| |ruffle membrane| |guanyl-nucleotide exchange factor activity| |response to calcium ion| |lipid binding| |cellular response to metal ion| |cellular response to inorganic substance| |Ras protein signal transduction| |synapse| |small GTPase mediated signal transduction| |neuron projection| |response to metal ion| |positive regulation of GTPase activity| |regulation of cell growth| |regulation of GTPase activity| |response to inorganic substance| |cell junction| |regulation of growth| |calcium ion binding| |positive regulation of hydrolase activity| |regulation of hydrolase activity| |positive regulation of catalytic activity| |intracellular signal transduction| |positive regulation of molecular function| \\ === CRISPR Data === ^Screen^Score^ |Oligomycin-A 20μM R00 exp26|-2.05| |LLY-283 2.6μM R06 exp290|-1.85| |BI-D1870 3.15μM R02 exp66|-1.71| |Ethambutol 25μM R08 exp481|1.93| No correlation found to any other genes in chemogenomics.
