The encoded protein is involved in transcription regulation and chromatin remodeling. Defects in this gene are associated with isolated cleft palate and cognitive disability. Alternate splicing results in multiple transcript variants that encode the same protein. [provided by RefSeq, Feb 2010]. * **UniProt Summary**: Binds to DNA, at nuclear matrix- or scaffold-associated regions. Thought to recognize the sugar-phosphate structure of double-stranded DNA. Transcription factor controlling nuclear gene expression, by binding to matrix attachment regions (MARs) of DNA and inducing a local chromatin-loop remodeling. Acts as a docking site for several chromatin remodeling enzymes and also by recruiting corepressors (HDACs) or coactivators (HATs) directly to promoters and enhancers. Required for the initiation of the upper- layer neurons (UL1) specific genetic program and for the inactivation of deep-layer neurons (DL) and UL2 specific genes, probably by modulating BCL11B expression. Repressor of Ctip2 and regulatory determinant of corticocortical connections in the developing cerebral cortex. May play an important role in palate formation. Acts as a molecular node in a transcriptional network regulating skeletal development and osteoblast differentiation. {ECO:0000269|PubMed:14701874}. |Homeobox| |CUT| |commitment of neuronal cell to specific neuron type in forebrain| |forebrain neuron fate commitment| |osteoblast development| |histone deacetylase complex| |forebrain neuron differentiation| |embryonic pattern specification| |neuron fate commitment| |forebrain generation of neurons| |roof of mouth development| |embryonic skeletal system morphogenesis| |nuclear matrix| |neuron migration| |embryonic skeletal system development| |osteoblast differentiation| |chromatin remodeling| |cartilage development| |central nervous system neuron differentiation| |transcription factor complex| |connective tissue development| |skeletal system morphogenesis| |cell fate commitment| |ossification| |embryonic organ morphogenesis| |RNA polymerase II regulatory region sequence-specific DNA binding| |forebrain development| |chromatin binding| |embryonic organ development| |pattern specification process| |DNA-binding transcription activator activity, RNA polymerase II-specific| |skeletal system development| |RNA polymerase II proximal promoter sequence-specific DNA binding| |embryonic morphogenesis| |chordate embryonic development| |embryo development ending in birth or egg hatching| |chromatin organization| |brain development| |head development| |negative regulation of transcription by RNA polymerase II| |animal organ morphogenesis| |cell migration| |embryo development| |central nervous system development| |neuron differentiation| |chromosome organization| |cell motility| |localization of cell| |negative regulation of transcription, DNA-templated| |positive regulation of transcription by RNA polymerase II| |negative regulation of nucleic acid-templated transcription| |negative regulation of RNA biosynthetic process| |locomotion| |negative regulation of RNA metabolic process| |negative regulation of cellular macromolecule biosynthetic process| |negative regulation of nucleobase-containing compound metabolic process| |negative regulation of macromolecule biosynthetic process| |negative regulation of cellular biosynthetic process| |generation of neurons| |positive regulation of transcription, DNA-templated| |negative regulation of biosynthetic process| |movement of cell or subcellular component| |DNA-binding transcription factor activity, RNA polymerase II-specific| |neurogenesis| |positive regulation of nucleic acid-templated transcription| |positive regulation of RNA biosynthetic process| |cell development| |negative regulation of gene expression| |positive regulation of RNA metabolic process| |tissue development| |positive regulation of nucleobase-containing compound metabolic process| |positive regulation of macromolecule biosynthetic process| |positive regulation of cellular biosynthetic process| |positive regulation of gene expression| |positive regulation of biosynthetic process| \\ === CRISPR Data === ^Screen^Score^ |THZ531 0.11 to 0.125 to 0.35μM on day4 then day6 R07 exp412|-2.26| |Dorsomorphin 5μM R07 exp357|-2.16| |Cytochalasin-B 5μM R07 exp349|-2.13| |FK-506 30μM R07 exp359|-2.04| |Lead acetate 2000μM R08 exp498 no dilution day6|-2.03| |CCT251545 20μM R07 exp326|-2.02| |Fluvastatin 2.2μM R06 exp282|-1.93| |Lead acetate 2000μM R08 exp497|-1.93| |2-Methoxyestradiol 0.2μM R01 exp40|-1.92| |Olaparib 4μM R08 exp512|-1.87| |THZ531 0.11 to 0.125μM on day4 R07 exp410|-1.81| |Parthenolide 2.5μM R05 exp238|-1.8| |Latrunculin-B 10μM R07 exp374|-1.77| |All-trans-Retinoic-Acid 40μM R07 exp334|-1.76| |Combretastatin A4 0.002 to 0.003μM day4 R04 exp179|-1.75| |CCCP 1μM R00 exp11|-1.73| |BN82002 4μM R07 exp340|-1.7| |CAY10603 0.55μM R08 exp467|1.76| |Thapsigargin 0.005μM R07 exp407|1.78| No correlation found to any other genes in chemogenomics.
