The encoded protein may be best known for its role in the trafficking and activation of prohormone convertase PC2 (encoded by Gene ID: 5126). Phosphorylation of the encoded protein has been shown to have an inhibitory effect on its chaperone function. [provided by RefSeq, Jul 2016]. * **UniProt Summary**: Acts as a molecular chaperone for PCSK2/PC2, preventing its premature activation in the regulated secretory pathway. Binds to inactive PCSK2 in the endoplasmic reticulum and facilitates its transport from there to later compartments of the secretory pathway where it is proteolytically matured and activated. Also required for cleavage of PCSK2 but does not appear to be involved in its folding. Plays a role in regulating pituitary hormone secretion. The C-terminal peptide inhibits PCSK2 in vitro. {ECO:0000269|PubMed:7913882}. |Secretogranin V| |enzyme inhibitor activity| |peptide hormone processing| |secretory granule| |neuropeptide signaling pathway| |unfolded protein binding| |protein processing| |hormone metabolic process| |protein maturation| |regulation of hormone secretion| |GTP binding| |peptide metabolic process| |regulation of hormone levels| |regulation of secretion by cell| |cellular amide metabolic process| |negative regulation of catalytic activity| |regulation of secretion| |intracellular protein transport| |negative regulation of molecular function| |proteolysis| |G protein-coupled receptor signaling pathway| |protein transport| |intracellular transport| |peptide transport| |amide transport| |cellular protein localization| |cellular macromolecule localization| |establishment of protein localization| |establishment of localization in cell| |nitrogen compound transport| |regulation of transport| |extracellular region| |gene expression| \\ === CRISPR Data === ^Screen^Score^ |Heregulin-B 44ng/ml R08 exp486|1.7| |ABT-702 5μM plus Deferoxamine 11μM R07 exp321|1.88| ^Gene^Correlation^ |RRM1|0.409| Global Fraction of Cell Lines Where Essential: 0/739 ^Tissue^Fraction Of Cell Lines Where Essential^ |1290807.0|0/1| |909776.0|0/1| |bile duct|0/28| |blood|0/28| |bone|0/26| |breast|0/33| |central nervous system|0/56| |cervix|0/4| |colorectal|0/17| |esophagus|0/13| |fibroblast|0/1| |gastric|0/16| |kidney|0/21| |liver|0/20| |lung|0/75| |lymphocyte|0/16| |ovary|0/26| |pancreas|0/24| |peripheral nervous system|0/16| |plasma cell|0/15| |prostate|0/1| |skin|0/24| |soft tissue|0/9| |thyroid|0/2| |upper aerodigestive|0/22| |urinary tract|0/29| |uterus|0/5| == Essentiality in NALM6 == * **Essentiality Rank**: 8668 * **Expression level (log2 read counts)**: 1.27
